反相HPLC法同时测定复方黄连素片中的8种成分

摘　　要：目的建立反相HPLC法同时测定复方黄连素片（盐酸小檗碱、木香、吴茱萸、白芍）中芍药内酯苷、芍药苷、盐酸小檗碱、吴茱萸内酯、吴茱萸碱、吴茱萸次碱、木香烃内酯和去氢木香内酯的方法。方法复方黄连素片提取物的定量分析采用月旭XB-C18色谱柱（250 mm×4.6 mm,5μm）;流动相为乙腈（A）-0.03 mol/L磷酸二氢钾水溶液（B）,梯度洗脱;体积流量1.0 m L/min;柱温25℃;检测波长220 nm。结果芍药内酯苷、芍药苷、盐酸小檗碱、吴茱萸内酯、吴茱萸碱、吴茱萸次碱、木香烃内酯和去氢木香内酯分别在0.054~0.804μg、0.080~1.21μg、0.132~1.99μg、0.034~0.504μg、0.035~0.519μg、0.031~0.468μg、0.048~0.714μg、0.053~0.792μg范围内与色谱峰面积呈良好的线性关系,加样回收率（n=6）分别为100.2%、99.2%、99.5%、99.2%、98.8%、100.5%、98.6%、99.0%,RSD分别为1.7%、1.5%、1.2%、1.9%、2.0%、1.9%、1.8%、1.0%。结论在80%甲醇溶液和超声30 min的提取条件下,该方法适用于复方黄连素片中这8个成分的测定。AIM To establish a method for simultaneously determining albiflorin,paeoniflorin,berberine hydrochloride,evodine,evodiamine,rutaecarpine,costunolide and dehydrocostuslactone in Compound Huangliansu Tablets（ berberine hydrochloride,Aucklandiae Radix,Evodiae Fructus,Paeoniae Radix Alba） by RP-HPLC.METHODS The quantitative analysis of Compound Huangliansu Tablets extract was carried out on Yue Xu XBODS C18column（ 250 mm × 4. 6 mm,5 μm）,mobile phase was acetonitrile（ A）-0. 03 mol / L potassium dihydrogen phosphate aqueous solution（ B） with gradient elution,flow rate was 1 m L / min,column temperature was maintained at 25 ℃,and detection wavelength was set at 220 nm. RESULTS The linear relationships of albiflorin,paeoniflorin,berberine hydrochloride,evodine,evodiamine,rutaecarpine,costunolide and dehydrocostuslactone fell within the ranges of 0. 054- 0. 804 μg,0. 080- 1. 21 μg,0. 132- 1. 99 μg,0. 034- 0. 504 μg,0. 035- 0. 519 μg,0. 031- 0. 468 μg,0. 048- 0. 714 μg and 0. 053- 0. 792 μg,respectively. Their average recoveries（ n = 6） were 100. 2%,99. 2%,99. 5%,99. 2%,98. 8%,100. 5%,98. 6% and 99. 0% with the relative standard deviations（ RSDs） of 1. 7%,1. 5%,1. 2%,1. 9%,2. 0%,1. 9%,1. 8%,and 1. 0%,respectively.CONCLUSION The application of 80% methanol solution and ultrasonic extraction for 30 min in this method is suitable for the determination of these eight constituents in Compound Huangliansu Tablets.
