摘  要:目的建立测定大鼠血浆中山姜素和小豆蔻明含有量的UHPLC-ESI-MS/MS法,并计算两者的药动学参数。方法血浆分析采用PhenomenexLuna C18色谱柱(150 mm×2.0 mm,3μm);流动相为乙腈-0.1%甲酸水溶液(20∶80);体积流量为0.4 m L/min;山姜素和小豆蔻明均采用正离子模式检测。结果标准品在1~1 000 ng/m L范围内线性关系良好,相关系数大于0.995,山姜素和小豆蔻明的最低检出限均为0.05 ng/m L,最低定量限均为0.2ng/m L,样品回收率在88.2%~99.3%之间。大鼠灌胃草豆蔻提取物后,两者的主要药动学参数Cmax分别为(385.633±91.192)和(186.683±69.893)ng/m L,t1/2z分别为(1.578±0.239)和(1.137±0.385)h,AUC(0-t)分别为(911.723±59.208)和(456.043±23.99)ng/m L/h,Vz/F分别为(24.295±6.858)和(35.606±12.887)L/kg。结论山姜素和小豆蔻明在大鼠体内的吸收和消除均比较迅速。AIM To establish a method for determining the contents of alpinetin and cardamonin in rat plasma by UHPLC-ESI-MS / MS and calculate their pharmacokinetic parameters. METHODS The analysis of plasma was performed on Phenomenex Luna C18column( 150 mm × 2. 0 mm,3 μm) with acetonitrile-0. 1% formic acid aqueous solution( 20 ∶ 80) as mobile phase,flow rate was 0. 4 m L / min,both alpinetin and cardamonin were detected in positive ion mode. RESULTS Calibration curves offered good linearities in the ranges of 1 ~ 1 000 ng / m L( r 0. 995). The detection limits of both alpinetin and cardamonin were 0. 05 ng / m L,while their quantitation limits were both 0. 2 ng / m L. The recoveries of samples were in the ranges of 88. 2% ~ 99. 3%. The main pharmacokinetic parameters of alpinetin and cardamonin in rats after intragastric administration were that Cmaxwere( 385. 633 ± 91. 192) and( 186. 683 ± 69. 893) ng /m L,t1 /2zwere( 1. 578 ± 0. 239) and( 1. 137 ± 0. 385) h,AUC( 0- t)were( 911. 723 ± 59. 208) and( 456. 043 ± 23. 99) ng / m L / h, Vz/ F were( 24. 295 ± 6. 858) and( 35. 606 ± 12. 887) L/kg,respectively. CONCLUSION Both alpinetin and cardamonin are rapidly absorbed and eliminated in rats.
