摘 要:目的建立GC-MS法比较铁皮石斛试管苗不同部位中挥发油的成分。方法正己烷-水蒸气蒸馏法提取铁皮石斛试管苗根、茎和叶中的挥发油,再利用GC-MS联用仪分析鉴定其化学成分和含有量。结果共鉴定出54种化学成分,其中根中16种、茎中45种、叶中10种,而且均以有机酸(亚油酸、棕榈酸等)为主,分别占挥发油总成分的64.40%、53.48%和96.04%。其中,根中依次为酯类(12.48%)、萜烯类(4.31%)、烷烃类(4.13%)和醇类(2.41%),其他占12.18%;茎中依次为醇类(20.66%)、萜烯类(12.27%)、酯类(6.07%)和烷烃类(2.26%),其他占5.25%;叶中其他类成分仅占6.02%。结论根和茎有9种共有成分,根和叶有7种共有成分,茎和叶有5种共有成分,但根、茎和叶仅有4种共有成分。而且,茎中存在多种根、叶中不含有的成分,主要为萜烯类和烯醇类化合物。AIM To establish a method to compare volatile oils in different parts of Dendrobium officinale tube seedlings by GC-MS. METHODS The volatile oils in roots,stems and leaves of D. officinale tube seedlings were obtained by n-hexane-distillation,whose chemical constituents and contents were then analyzed by gas chromatography-mass spectrum( GC-MS). RESULTS A total of 54 chemical constituents were identified,45 of which were found in stems,16 of which were found in roots,10 of which were found in leaves. And most of them were organic acids such as inoleic acid and hexadecanoic acid,accounting for 64. 40%,53. 48% and 96. 04% in roots,stems and leaves, respectively. Among them, roots were followed by esters( 12. 48%), terpenes( 4. 31%),alkanes( 4. 13%),alcohols( 2. 41%) and others( 12. 18%),stems were followed by alcohols( 20. 66%),terpenes( 12. 27%),esters( 6. 07%),alkanes( 2. 26%) and others( 5. 25%),while other chemical constituents only accounted for 6. 02% in leaves. CONCLUSION There are 9 common constituents in roots and stems. Seven constituents exist in roots and leaves. There are 5 constituents in stems and leaves. Only 4 constituents exist in roots,stems and leaves. In addition,many constituents exist in stems that don't exist in roots and leaves. They are mainly terpene and enol compounds.
