摘 要:目的探讨人脐带血间充质干细胞(human umbilical cord mesenchymal stem cell,HUCMSC)在体内外对类风湿性关节炎(rheumatoid arthritis,RA)的作用效果,为RA的干细胞治疗提供新的依据。方法建立胶原诱导类风湿性关节炎(collageninduced arthritis,CIA)小鼠模型,连续5 d每天腹腔注射100μl PBS(对照组)或100μl PBS与HUCMSC(5×106/100μl)的混合液,从治疗第1天起每周3次,监测CIA小鼠关节发病临床评分变化以及体质量变化,直至实验结束。结果 HUCMSC可减轻CIA小鼠关节发病程度,抑制促炎因子肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、白介素1β(interleukin-1β,IL-1β)和IL-6的生成。结论 HUCMSC可减轻和缓解RA的临床症状,抑制其发生和发展,为研究和治疗RA提供了新思路。This study performed to explore the therapeutic effect of human umbilical cord mesenchymal stem cell(HUCMSC) in treating rheumatoid arthritis(RA), and supply a new evidence for HUCMSC treatment of RA. A collagen-induced arthritis mouse model was built, and injected with 100 μl PBS(control group) or HUCMSC(5×106/100 μl) plus 100 μl PBS(experimental group) continuously through abdominal cavity. Then, the body weight and joint clinical score changing were monitored three times every week from the day treatment. Finally, the changing of mouse's joint and proinflammatory cytokines were analyzed. Data showed that HUCMSC could alleviate mouse joint pathology status and inhibit tumor necrosis factor-α, interleukin-1β and IL-6 production. In conclusion, HUCMSC could relief RA clinical symptom and inhibit RA development, which provides a new idea to study and treat RA.
