摘 要:目的探讨哮喘患者体内IL-35与CD8+T细胞之间的关系及IL-35在哮喘患者体内可能的作用。方法采集哮喘患者和正常人外周静脉血,分离、培养外周血单个核细胞(PBMCs);流式细胞术检测Tregs、Tc1、Tc2和Tc17细胞比例;ELISA检测血浆IL-35、IFN-γ、IL-4和IL-17水平。采用Pearson检验分析哮喘患者血浆IL-35与Tc1、Tc2和Tc17细胞及其相关细胞因子之间的关系。结果与正常人相比,哮喘患者外周血Tregs细胞及IL-35表达水平明显下降(P<0.01);Tc2和Tc17细胞及相关细胞因子(IL-4、IL-17)显著增高(P<0.05);血浆IL-35水平与外周血Tc2细胞数目及IL-4水平成明显负相关性(P<0.01)。结论哮喘患者血浆IL-35与外周血分泌IL-4的CD8+T(Tc2)细胞成负相关,体内降低的IL-35可能参与了哮喘患者的发病过程。Interleukin(IL)-35 is a newly discovered suppressive cytokine and has been shown to alleviate inflammatory and autoimmune diseases. The purpose of this study was to analyze the correlations between interleukin-35(IL-35) levels and CD8+T cells, and investigate potential immunomodulatory capacity of IL-35 in asthmatic patients. Heparinized venous blood was collected from asthmatic patients and healthy controls. Peripheral blood mononuclear cells(PBMCs) were isolated by Ficoll gradient centrifugation and cultured in vitro. The frequencies of cytotoxic T cells(Tc)1, Tc2, Tc17 and Tregs were measured by flow cytometry. Plasma levels of IL-35, interferon(IFN)-γ, IL-4, and IL-17 were examined by enzyme-linked immunosorbent assay(ELISA). The correlations between plasma IL-35 levels and Tc1, Tc2, or Tc17 cytokine production in asthmatics were analyzed by Pearson's test. Data showed that the frequency of Tregs and IL-35 protein levels were down-regulated in asthmatic patients compared with healthy controls(P 0.01). While the frequencies of Tc2 and Tc17 cells were significantly increased in asthmatic patients(P 0.05). Similarly, plasma levels of IL-4 and IL-17 were significantly increased in asthmatics(P 0.05). More importantly, plasma IL-35 protein levels were negatively correlated with the frequency of Tc2 cells and the level of IL-4 in asthmatic patients(P 0.01). Our results suggest that decreasing of circulating IL-35 levels could contribute to the pathogenesis of asthmatic patients.
