摘 要:目的超声波法优化菊花叶总黄酮提取工艺。方法以芦丁为对照,在单因素试验的基础上以乙醇浓度、料液比、超声时间、超声功率为考察因素,利用紫外分光光度计测定总黄酮吸光度,以总黄酮含量为评价指标,采用L9(34)正交实验优化提取工艺,运用D101大孔吸附树脂分离富集菊花叶提取液中总黄酮。结果菊花叶总黄酮的最佳提取工艺为:80%乙醇溶液为提取溶剂,采用料液比1:25g/m L,在250w超声功率下超声25min;且D101大孔吸附树脂能较好分离富集菊花叶黄酮类物质。结论提取工艺方法简单、合理、稳定且可行,该研究为菊花叶中有效成分的综合利用提供了实验基础。Objective optimizing the extraction process of total flavonoids from forists chrysan- themum leaf. Methods Using rutin as reference substance, the extract technology of the total flavonoids from florists chrysanthemum leaf was optimized by orthogonal experiment with the yield of total flavonoids from florists chrysanthemum leaf as index and with the concentration of ethanol, the ratio of solid to liquid, the ultrasonic time, the ultrasonic power as factors. Flavonoid was determined by ultraviolet spectrophotometer. D101 types of macro reticular adsorbents were used to separate and purify total flavones from extracting solution of florists chrysanthemum leaf. Results The optimum process of the extraction was that the superior technology of total flavonoids was as follows: 80% alcohol, 25 - fold solvent amount, 25 minutes and the ultrasonic power is 250w. Conclusion Experiment results made it clear that because the rate of total flavonoids extracted from florists chrysanthemum leaf by the optimum extracting technology was simple, reasonable, stable and practicable, D101 types of macro reticular adsorbents could separate and purify total flavones of the florists chrysanthemum leaf well.
