摘  要:目的为了调控纯镁的医用活性,将纯镁超声微弧氧化涂层进行化学镀锌,研究其对成骨细胞增殖和分化的影响。方法将纯镁超声微弧氧化后,进行化学镀锌,制备出表面含锌的涂层材料。实验分为三组:A组为超声微弧氧化对照组(MAO),B组为化学镀锌实验组(MAO-Zn),C组为碱处理后化学镀锌实验组(MAO-Na OH-Zn),用扫描电镜(SEM)测量与研究各组试件表面的形态,激光共聚焦显微镜观察成骨细胞的早期形态,用CCK-8法检测细胞增殖,并检测成骨细胞ALP活性。结果扫描电镜和激光共聚焦显示C组和B组表面细胞的生长和黏附均优于A组,C组优于B组。CCK-8和ALP的检测结果显示三组材料表面细胞的增殖、活性为C组>B组>A组,三组的比较差异均具有统计学意义。结论含锌MAO涂层具有良好的生物相容性,而经碱处理后的镀锌涂层生物相容性最佳。Objective Investigate the effect from pure magnesium to osteoblasts' proliferation and differentiation,which is coated with zinc on its ultrasonic micro-arc oxidation( MAO) for controlling its medical activity. Methods To prepare the zinc coating on pure magnesium by MAO and create coating material containing zinc on the surface. The experiment was divided into three groups: group A was the control group of ultrasonic micro-arc oxidation( MAO),group B was the experiment group of zinc( MAO-Zn),and group C was the experiment group of zinc after alkali treatment( MAO-Na OH-Zn). To measure and analyze the surface morphology of the test-pieces by scanning electron microscopy( SEM),observe the early morphology of osteoblasts by confocal laser scanning microscopy( CLSM). To detect the proliferation and the ALP activity of the cells by CCK-8. Results The CLSM results showed that the growth and adherence of cells in the groups C and B was better than group A,and the group C was better than group B,while the results from CCK-8 and ALP assays showed that the proliferation / viability of the cells in the 3groups were group C group B group A( P 0. 05 for all). Conclusions The pure magnesium coated with zinc by MAO possesses good biocompatibility,and the alkali treated material is the best.
