摘  要:目的:研究祛白片对白癜风小鼠模型的疗效及其相关作用机制。方法:采用5%H2O2建立小鼠白癜风模型,实验设空白对照组、模型对照组和高、中、低剂量给药组。肉眼观察各组小鼠皮肤色泽变化及其毛发生长情况,同时采用HE染色法,镜下观察皮肤组织形态学改变。采用全基因芯片测试方法探讨祛白片的作用机制。结果:整体动物实验结果显示实验高、中、低剂量组与模型组相比疗效有统计学差异,各剂量组间具有浓度依赖性。HE染色结果显示,与模型组相比,高、中剂量组小鼠皮肤受试区出现大量黑色素颗粒,同时毛囊数恢复。小鼠全基因芯片研究结果显示,给药组与模型组相比共2 219个基因转录有变化,这些差异基因分别对文献报导的7种白癜风发病机制中的4种机制有调控作用。初步确认祛白片的作用机制与抑制自身免疫系统亢进、促进角质细胞形成、降低氧化应激反应和促进黑色素形成相关。结论:本研究证实了祛白片的抗白癜风作用,同时对其分子机制进行了初步探索,为后期深一步研究提供方向。OBJECTIVE To study effects and mechanism of caltrop complex on mice bearing vitiligo. METHODS Five percent hydrogen peroxide solution was adopted to scrub skin of experimental mice to establish vitiligo mouse model. Then animals were divided into 5 groups randomly. Effects were evaluated and mouse genome oligo microarray was adopted to study the mechanisms. RESULTS Compared with model group, effects were statistically different between high, middle and low dose groups. HE staining showed a large number of melanin granules visible in high and middle dose groups. Gene chip results showed that transcriptions of 2 219 genes were changed and mechanisms of caltrop complex might be focused on autoimmune system, keratinocytes formation, oxidative stress and melanin biosynthesis. CONCLUSION Caltrop complex can improve pigmentation in tested skin areas and make black hair growing. Multiple pathways are affected by this agent to promote skin from vitiliginous to black.
