摘 要:目的 比较研究不同条件下体外扩增的造血细胞回输体内后促进造血功能恢复作用的差异 ,方法 在小鼠骨髓单个核细胞液体培养体系中 ,分别加入几种细胞因子组合和 /或基质细胞层存在的条件下进行体外扩增 ,然后将不同条件下扩增的细胞经尾静脉回输至经致死量照射的小鼠体内 ,动态观察小鼠的外周血象变化及其一般状况和生存率。结果 ①单纯细胞因子介导BMMNC体外扩增后并不能增进这些细胞促造血恢复的能力 ;②含有骨髓基质细胞底层的体外扩增 ,无论是否加入细胞因子 ,均有明显的促进移植受体造血功能迅速恢复的作用。结论 骨髓基质细胞支持下的造血细胞体外扩增可能有助于维持扩增后造血干 /祖细胞造血重建的功能 。Objective To compare the effects of ex vivo expanded bone marrow cells under different conditions on hematopoietic repair of irradiated mice. Methods The mouse bone marrow mononuclear cells (BMMNC) were expanded for 5 d in the liquid expanded cultural system with several cytokines and /or bone marrow stromal cells layer and the expanded cells were transplanted to the lethal dose irradiated mice respectively via the caudal vein. The hematopoietic recovery of mice after transplantation was evaluated by analyzing the peripheral blood Hb, WBC, and TBC, and by observing the state and survival rates of the mice. Results Ex vivo expansion of BMMNC under the cultural conditions with combined cytokines didn't improve the hematopoietic recovery of irradiated mice, but the expansion with the aid of bone marrow stromal cells accelerated this process significantly. Conclusion The results suggest that the ex vivo hematopoietic cells expansion supported by bone marrow stromal cells can maintain the properties of the hematopoietic stem/progenitor cells.
