灵芪胶囊对直结肠癌lovo细胞凋亡相关基因表达的影响

摘　　要：目的:观察灵芪胶囊对直结肠癌lovo细胞凋亡相关Caspase3基因表达的影响。方法:灵芪胶囊含药血清根据浓度不同分为灵芪胶囊高剂量组、灵芪胶囊中剂量组、灵芪胶囊低剂量组、阳性对照组及空白对照组,将对数生长期的lovo细胞培养至细胞贴壁后,分别放入相应的含药血清,48 h后收集细胞,应用蛋白质印迹法检测各组Caspase3的含量。结果:空白对照组Caspase3蛋白表达量较高,阳性对照组和灵芪胶囊高剂量组Caspase3蛋白表达量小于空白对照组,其结果具有明显统计学差异(P<0.01)。灵芪胶囊中剂量组、灵芪胶囊低剂量组Caspase3表达量与空白对照组相比,差异有统计学意义(P<0.05)。结论:体外条件下,灵芪胶囊对lovo细胞具有一定的抑制作用,能够使细胞凋亡发生。灵芪胶囊导致细胞发生凋亡的作用机制可能与抑制Caspase3基因的表达情况有关。Objective： To study the expressive effect on the apoptosis-related genes with colorectal cancer lovo cell by Lingqi capsules.Methods： Lingqi capsules were divided into high dose group,middle dose group,low dose group,positive control group and blank control group,according to the medicated serum concentration. After culturing the logarithmic phase of lovo cell to cell adherence,into the corresponding drug-containing serum respectively. Collecting cells after 48 h. Protein imprinting method was applied to detect the content of each Caspase3 Results： High Caspase3 protein expression in the blank control group,positive control group and Lingqi capsules highdose group Caspase3 protein expression quantity is less than the blank control group,the result has significant statistical difference（ P〈0. 01）. Middle dose and low groups of the Lingqi capsules Caspase3 compared with the blank control group,the difference was statistically significant（ P〈0. 05）. Conclusion： Lingqi capsules on lovo cell in vitro condition has certain inhibitory effect,can make the cell apoptosis. Lingqi capsules in the mechanism of action of cell apoptosis may be associated with caspase3 gene expression,inhibit caspase3 to inhibit the action of the straight colorectal cancer.
