摘  要:目的:比较地黄叶、桑叶单用与合用对四氧嘧啶诱导的糖尿病模型小鼠降低血糖的作用。方法:KM雄性小鼠,禁食不禁水16 h后,以0.01 m L·g-1体质量的剂量腹腔注射新鲜配制的四氧嘧啶生理盐水溶液(23 g·L-1),建立小鼠糖尿病模型,造模成功后将造模组动物随机分为模型组、地黄叶组、桑叶组、地黄叶桑叶合用(1∶1)组、阳性对照(二甲双胍)组,各组灌胃给予相应药物10 d,模型组给予等体积的蒸馏水,余下各组剂量分别为:6 g·kg-1,6 g·kg-1,6 g·kg-1,6 g·kg-1,6 g·kg-1,0.2 g·kg-1。给药后观察各组小鼠的行为表现,以及摄食量、饮水量、体质量的变化。通过测定空腹血糖,观察地黄叶、桑叶单用与二者合用对糖尿病动物模型血糖的影响。结果:造模后小鼠平均体质量显著降低(P<0.01);饮水量和摄食量均显著增加(P<0.01);垫料潮湿,符合糖尿病小鼠"三多一少"的典型症状,且空腹血糖浓度超过11.1 mmol·L-1(P<0.01)。实验期间模型对照组小鼠血糖一直保持较高水平,未出现自行降低。灌胃给药后,各组小鼠的体质量有不同程度的增加,饮水量、摄食量、血糖值有不同程度的降低。结论:地黄叶桑叶合用的效果不如单用的效果好,地黄叶的降低血糖作用显著优于桑叶,与盐酸二甲双胍类似。Objective: To compare effects of Rehmannia glutinosa Libosch leaves and Folium mori administered alone and in combination on the diabetic model mouse induced by alloxan. Methods: KM male mice,fasted for 16 h after alloxan,saline solution with 1 mL /100 dose of g body weight intraperitoneal injection of freshly prepared( 23 mg · mL- 1),the establishment of diabetic mouse model. After modeling,model mice were randomly divided into model compared group,Rehmannia glutinosa Libosch leaves group,Folium mori group,1: 1 group and metformin hydrochloride group. Every group was given corresponding drugs for 10 days. The dose was 6 g·kg- 1,6 g·kg- 1,6 g·kg- 1,6 g·kg- 1,6 g·kg- 1,0. 2 g·kg- 1. 10 days laterig,weight,food intake,the amount of water,hypoglycemic and hyperlipemia were observe. Through the determination of fasting blood glucose,effects of Rehmannia glutinosa Libosch leaves and Folium mori administered alone and in combination on the diabetic model mouse. Results: After modeling,the average weight of mice was reduced significantly( P〈0. 01),the amount of drinking water and food intake was significantly increased( P〈0. 01),litter moisture,consistent with the typical symptoms of DM mice " three little",and the fasting blood glucose concentration is more than 11. 1 mmol·L- 1( P〈0. 01). During the experiment,the model control group mice blood glucose maintained high level,does not appear to reduce.After intragastric administration,all the mice weight increased in different degrees,the amount of water,food intake,blood glucose levels decreased in different degree. Conclusion: The combination of Rehmannia glutinosa Libosch leaves and Folium mori is worse than single using,the hypoglycemic effect of Rehmannia glutinosa Libosch leaves significantly,better than Folium mori( P〈0. 05),similar to metformin hydrochloride( P〉0. 05).
