摘 要:目的探讨益肾养阴合剂对肾功、肾脏病理变化及对P-STAT3表达的影响,揭示其发挥疗效的作用机制,为益肾养阴合剂治疗狼疮性肾炎(LN)提供理论依据。方法两种种系相同不同品种的50只小鼠随机分5组:正常组C57BL/6J小鼠(0.9%生理盐水);MRL/1pr小鼠根据尿蛋白含量随机分为4组:模型组、中药组、中药联合激素组、激素组,连续给药4周后观察P-STAT3、尿蛋白含量、血肌酐、尿素氮及肾脏病理变化。结果在给药第一周,与模型组相比,尿蛋白含量在各治疗组之间无明显差异(P>0.05)。在第2、3、4周时,与模型组相比,各治疗组的尿蛋白、血肌酐、尿素氮含量均有显著下降(P<0.05),中药联合激素组的尿蛋白含量最低(P<0.05)。给药处理后,p-stat3的表达均呈下降趋势,中药组、激素组的p-stat3表达相比模型组有所减少(P<0.05);中药联合激素组的降低情况最为显著(P<0.05)。HE染色三个治疗组肾组织局部轻度系膜细胞增生,肾小球炎症细胞浸润减少。结论 1.益肾养阴合剂能改善MRL/lpr小鼠的肾功能及肾脏病理,且联合激素肾功能改善更明显。2.下调磷酸化的STAT3的表达可能是益肾养阴合剂治疗狼疮性肾炎的作用靶点之一。Objective: To study the effect ofYishen Yangyin Mixture on renal function, renal pathological changes and P -STAT3 presentation and reveal its clinical mechanism in order to provide a theoretical basis for the treatment of lupus nephritis (LN). Methods : Two germ lines of different kinds of 50 mice were randomly divided into five groups. Normal C57BL/6J mice (0.9% saline) and MRL/1 pr mice were randomly divided into four groups according to urinary protein content, a model group, a TCM group, a TCM combined hormone group and a hormone group and were administrated for 4 weeks to observe their P - STA33, urine protein, serum creatinine, blood urea ni- trogen and renal pathological changes. Results: In the 1st week administration, compared with the model group, the urine protein of each group was no significant difference (P 〉 0.05). In the 2nd, 3rd and 4th week administration, compared with the model group, the urine protein, serum creatinine and urea nitrogen of each group significantly decreased (P 〈 0.05), among which the urine protein of the TCM combined hormone group was lowest (P 〈 0.05 ). After the administration, p - stat3 presentation showed a downward trend. The p - star3 presentation of the TCM group and the hormone group decreased when compared with that of the model group ( P 〈 0.05 ). The p - stat3 of the TCM combined hormone group decreased most significantly (P 〈 0.05 ). HE staining of renal tissue of three treatment groups showed local mild mesangial cell proliferation and glomerular inflammatory cell infiltration reduction. Conclusions: A. Yishen Yangyin Mixture can improve the renal function and renal pathology of MRL/lpr mice, and improve more obviously when combined hormone. B. The reduced phosphorylation STAT3 may be one of the targets for the treatment of lupus nephritis.
