摘 要:目的实时荧光定量PCR检测黑参对S180肿瘤模型小鼠TNF-αmRNA表达的影响,应用分子生物学手段分析黑参抑制肿瘤的机制。方法采取腋下注射S180肿瘤细胞制得小鼠肿瘤模型,随机分成空白对照组、环磷酰胺组、消癌平组、红参高剂量组、红参中剂量组、红参低剂量组、黑参高剂量组、黑参中剂量组和黑参低剂量组。除空白对照组外,其他实验组均采取同时灌胃给药。测定小鼠给药前后体重、给药后瘤重,计算抑瘤率,利用IDTDNA设计引物,应用实时荧光定量PCR检测不同组TNF-α表达丰度。结果与空白对照组相比,所有实验组肿瘤质量都有所减少。除消癌平和环磷酰胺外,使用高剂量黑参和低、中剂量红参对肿瘤重量的抑制较为显著(P<0.01)。实时荧光定量PCR显示,同对照组及其他实验组相比,高剂量黑参组和低剂量红参组TNF-α表达量上调(P<0.01,P<0.05)。结论综上,使用高剂量黑参和低剂量红参能够提高小鼠TNF-α转录表达水平并减少肿瘤重量,其中高剂量黑参组效果更明显。Objective: To detect the effect of Black Ginseng on TNF -α mRNA presentation of S180 tumor -bearing mice by real - time fluorescence quantitative PCR and analyze the tumor inhibition effect of Black Ginseng by molecular biological method. Methods: Tumor - bearing mice models were made by armpit injection of S180 tumor cells and were randomly divided into a control group, a cyclophosphamide group, aXiaoaiping group, high, medium and low dose groups of Red Ginseng and high, medium and low dose groups of Black Ginseng. Except the control group, the other experimental groups were given intragatric administration. The mice weight in the pre - ad- ministration and the mice tumor weight in the post - administration were measured. Anti - tumor rate was calculated and IDTDNA primers were used. Real - time fluorescence quantitative PCR was used to detect different TNF - α presentations. Results : Compared with the con- trol group, the tumor mass in the experimental groups declined. Except theXiaoaiping group and the cyclophosphamide group, the high - doseBlack Ginseng group and low and medium - doseRed Ginseng groups showed more obvious inhibition effect on tumor weight ( P 〈 0. 01 ). Realtime PCR showed that when compared with the control group and other groups, the high -doseBlack Ginseng group and the low doseRed Ginseng group upregulated TNF - α presentation ( P 〈 0.01, P 〈 0. 05 ). Conclusion : The high - doseBlack Ginseng group and the low - doseRed Ginseng group can improve the TNF - α transcriptional presentation in mice and reduce tumor weight, among which the high- doseBlack Ginsenggroup is more effective.
