摘 要:本文采用三维荧光光谱结合化学计量学交替三线性分解(ATLD)方法,对中药鬼臼和人体体液中鬼臼毒素的含量和回收率进行测定,利用核一致诊断法对体系的组分数进行了估计。采用标准曲线法和二阶标准加入法得到的鬼臼中鬼臼毒素的含量分别为4.855%±0.14%和4.558%±0.05%,回收率为101.4%±1.2%,人体血浆和尿液中鬼臼毒素的回收率分别为101.4%±3.9%和99.4%±0.9%,得到的鬼臼毒素的解析光谱与真实光谱几乎完全重合。实验结果表明,该方法可用于干扰组分共存下中药鬼臼和人体体液中鬼臼毒素的快速、准确定量分析。同时,讨论了内滤光效应对定量测定鬼臼毒素的影响及其校正方法,吸光度校正后的定量测定结果更好。We herein reported a simple method for determination of podophyllotoxin in podophyllum and body fluids even in the presence of other uncalibrated interferents by using excitation-emission matrix(EEM)fluorescence coupled with second-order calibration method based on alternating trilinear decomposition(ATLD)algorithm.The core consistency diagnostic(CORCONDIA)test indicated the correct number of components.The obtained results showed that the contents of podophyllotoxin by standard curve and second-order standard addition method with component number N =2 were 4.855% ±0.14% and4.558%±0.05%,respectively.The average recovery was 101.4%±1.2%,and the average recoveries in human plasma and urine were 101.4% ±3.9% and 99.4% ±0.9%,respectively.The resolved spectra and real spectra of podophyllotoxin matched quite well with each other.At the same time,the influence of the inner filter effect in human plasma and urine on fluorescence intensity was investigated.The results indicated that the quantitative determination results were more satisfactory after ultraviolet correction.
