摘  要:目的 探讨DNA聚丙烯酰胺凝胶电泳 (PAGE)三种银染方法的特点及其适用性。方法 采用非变性PAGE垂直板对不同稀释度的 pBR 32 2MspⅠ分子量标准进行电泳分离 ,利用二种硝酸银染色法和一种银氨染色法染色 ,分析其灵敏度和噪声。结果 分子量Marker的所有条带都能染出灵敏度均为 2 5ng。在至少一个条带能染出的灵敏度方面 ,银氨染色法 ( 6 .2 5ng)低于其他两种方法。结论 时间方面Carlos等所建立的银染方法最快 ,仅需 2 0min。银氨法的噪声最低 ,结果易于保存。Objective According to the principle of increasing the sensitivity and reducing the background, three DNA silver staining methods on Polyacrylamide gels electrophoresis (PAGE) were analyzed and evaluated. Methods Serial diluted pBR 322 MspⅠ DNA molecular weight standard were separated by non-denature vertical PAGE. Two nitrate silver and one dimmine silver staining methods have been used. Sensitivity and background of those methods were evaluated. Results The sensitivity of all the band of pBR 322 MspⅠ Marker developed is coequally 25 ng. For the aspect of at least single band developed, the sensitivity of dimmine silver staining methods (6.25 ng) inferior to the others. Conclusion The methods developed by Carlos J. et al. is the fastest (20 minitus). The background of dimmine silver staining methods is the lowest, and it's results is convenient for preservation. While the methods developed by Brant J. et al. have no superiority to the others in application.
