摘 要:通过对云南剑川30个大绒鼠样本线粒体细胞色素b基因(Cyt b)和控制区(D-loop)序列研究,初步探讨剑川大绒鼠的遗传多样性及系统发生。结果表明,剑川地区大绒鼠Cyt b包含16个变异位点(占全序列的1.40%),其中转换3个,颠换0个,核苷酸多样性(Pi)为0.00269,单倍型多态度(Hd)0.876±0.041,共定义12个单倍型。D-loop全长987 bp,包含28个变异位点(占全序列的2.84%),其中转换5个,颠换1个,Pi为0.0055,Hd为0.924±0.029,共定义16个单倍型。Cyt b和D-loop单倍型系统树显示,30个样本形成两支,且D-loop序列中显示出的多样性水平可推测其种群比Clethrionomys种群更年轻。其基因的多样性水平可能与其分布范围和生态因子有关。To investigate the genetic diversity of Orient voles ( Eothenomys miletus) , DNA sequence variations in the mitochondrial cytochrome b gene and control region of the 30 individuals from Jianchuan, Yunnan Province were measured.For cytochrome b gene (Cyt b), 16 nucleotide sites were variable (1.40% in the full sequences), including 3 transitions and 0 transversions among 1140 base pairs (bp), the nucleotide diversity (Pi) was 0.0296, the haplotypes diversity (Hd) was 0.876 ±0.041, and 12 haplotypes were identified in the 30 samples.For control region (D-loop), 987 base pairs were examined, 28 nucleotide sites were variable (2.84%in the full sequences), including 5 transitions and 1 transversions, the nucleotide diversity was 0.0055, the haplotypes diver-sity was 0.924 ±0.029, 16 haplotypes were identified in the 30 samples.Based on control region, the genetic diversity of Eothenomys was less than Clethrionomys, so the Eothenomys could be younger than Clethrionomys, and the genetic diversity may be related with the distribution ranges and ecological factors.
