清脂胶囊抗大鼠营养性肥胖的药效与机制

摘　　要：目的:通过观察高、中、低剂量组的清脂胶囊对营养性肥胖大鼠各项指标的影响,进一步明确该药药效及最佳剂量,初步阐明该方抗营养性肥胖的机制。方法:采用高脂乳剂+碳酸饮料+普通饲料混合喂养复制大鼠营养性肥胖模型。各组动物末次给药后,检测血清葡萄糖(GLU)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、瘦素(LEP)、脂联素(ADP)、胰岛素(INS)含量,计算脂肪指数。结果:与模型对照组比较,清脂胶囊高、中剂量组可降低大鼠血清GLU、TG含量,清脂胶囊低剂量组可降低血清TG水平,而清脂胶囊中、低剂量组则显著升高血清HDL-C水平(P<0.05,P<0.01);清脂胶囊高剂量组可降低血清LEP、ADP、INS水平,清脂胶囊中、低剂量组可分别降低ADP、LEP水平(P<0.01)。结论:清脂胶囊高、中、低剂量组均可对抗混合喂养法所致大鼠营养性肥胖的发生,其作用可能与干预肥胖大鼠LEP抵抗,降低血清INS水平有关;综合药效和用药安全度,临床建议采用低剂量作为首选剂量。Objective：To know the pharmacological function and optimal dosage of Qingzhi Capsule and clarify the mechanisms of resistingnutritional obesity rat with this formula by observing the influence of high,middle and low dosage Qingzhi Capsule on indexes of rats with nutritional obesity.Methods：The rat models of nutritional obesity were established by feeding with mixture of high fat emulsion,carbonated beverage and general diet.The levels of GLU,TC,TG,LDL-C,HDL-C,LEP,ADP,and INS in serum of rats in each group were tested after the administration,and the fat index of rats was also calculated.Results：The high and middle dosage of Qingzhi Capsule could decrease the levels of GLU and TG in serum of rats,and the low dosage of Qingzhi Capsule could decrease serum TG level of rats.Middle and low dosage of Qingzhi Capsule could increase the levels of HDL-C（P〈0.05,P〈0.01）,and high dosage of Qingzhi Capsule could decrease the levels of LEP,ADP and INS in serum of rats.The middle dosage of Qingzhi Capsule could decrease ADP and the low dosage of Qingzhi Capsule could decrease LEP（P〈0.01）.Conclusion：The high,middle and low dosage of Qingzhi Capsule could be used to resistnutritional obese rats induced by mixed feeding,and the mechanism might related with the intervention effect in LEP resistance of nutritional obese rats and the effect on serum INS levels.Considering the efficacy and safety of Qingzhi Capsule,it suggests that the low dosage could be as the preferred dose in clinical.
