摘 要:目的:通过p38、p42/44 MAPK通路观察茵陈五苓散对高脂血症大鼠模型血脂的影响。方法:通过高脂饲料喂饲法复制高脂血症大鼠模型。将成模的SD大鼠随机分为模型组、茵陈五苓散组(治疗组)、血脂康组(对照组),分别灌胃0.9%氯化钠溶液、茵陈五苓散、血脂康;50d后检测大鼠血脂,并活检肝脏组织,提取总RNA,运用QPCR分析肝脏组织低密度脂蛋白受体(LDL-R)的表达;提取总蛋白,Western blotting分析p38MAPK、p42/44 MAPK表达和磷酸化水平,并对活检的肝脏组织切片进行免疫组化。结果:与模型组比较,茵陈五苓散组能够降低高脂血症模型大鼠的总胆固醇(TC)、甘油三酯(TG)及低密度脂蛋白(LDL-C)(P<0.01),升高高密度脂蛋白(HDL-C)(P<0.05),使LDL-R表达明显升高(P<0.01);同时抑制p38 MAPK磷酸化,提高p42/44 MAPK磷酸化水平(P<0.05)。结论:茵陈五苓散通过调节p38 MAPK和p42/44 MAPK及其磷酸化水平,而调控肝细胞膜上LDL-R的表达,治疗高脂血症。Objective:To observe the influence of Yinchen Wulin Powder on the blood fat of hyperlipidemia rat models via p38 and p42/44 MAPK pathway.Methods:Hyperlipidemia rat models were established by feeding with high-fat diet.After rat models establishing,rats were randomly divided into model group,Yinchen Wulin Powder group(treatment group) and Xuezhikang group(control group),rats in these groups received gavage administration with normal saline,Yinchen Wuling Powder and Xuezhikang respectively for 50 days,and then the blood fat of rats was tested and the liver tissue of rats was tested by using biopsy.The mRNA expression of LDL-R in liver of rats was tested by using qPCR,and the expression and phosphorylation level of p38 MAPK and p42/44 MAPK were tested by using Western blotting and immunohistochemistry.Results:Compared with model group,Yinchen Wuling Powder could decrease the levels of TC,TG and LDL-C,increase the levels of HDL-C and LDL-R,inhibit the phosphorylation of p38 MAPK,and improve phosphorylation level of p42/44 MAPK of hyperlipidemia rats simultaneously.Conclusion:Yinchen Wuling Powder could treat hyperlipidemia through regulating the expression of LDL-R of liver cell membrane,and adjusting the expression and phosphorylation level of p38 MAPK and p42/44 MAPK.
