摘 要:目的:观察VEGF、VEGFR-1、VEGFR-2在多个组织的表达,评价保肺定喘汤在COPD大鼠气道重塑中抗血管新生的作用。方法:30只雄性SD大鼠随机分为3组,其中,正常组大鼠不熏烟,不用药;中药组和模型组则选用气道内滴入脂多糖刺激法及熏烟法进行造模。造模成功后前者予保肺定喘汤2m L/d灌胃治疗,后者则予等量0.9%氯化钠溶液作为对照。28d后处死大鼠,行HE染色、免疫组化法及ELISA观察血清、BALF、肺组织中VEGF、VEGFR-1、VEGFR-2水平变化。结果:与模型组比较,中药组大鼠细支气管、肌性动脉形态及肺组织病理改变有所改善,且血清中VEGF、VEGFR-1、VEGFR-2及BALF、肺组织中VEGF、VEGFR-1表达减少(P<0.05)。结论:保肺定喘汤能改善COPD大鼠气道重塑,其机制可能与降低VEGF传导通路中的VEGF、VEGFR-1、VEGFR-2表达,抑制血管新生相关。Objective:To observe the antiangiogenic effect of Baofei Dingchuan Decoction in the airway remodeling process of COPD rats by observing the expression of VEGF,VEGFR-1 and VEGFR-2 inmultiple tissues of COPD rats.Methods:Thrity male SD rats were randomly divided into three groups.Rats in control group did not receive any stimulation and treatment.Rats in Chinese medicine group and model group received lipopolysaccharide and fumigation stimulations in tracheas to establish the rat models.After model establishing,rats in Chinese medicine group received gavage administration with Baofei Dingchuan Decoction(2mL/d),and rats in model group received gavage administration with normal saline(NS).After treating for 28 d,all the rats were sacrificed and the expression of VEGF,VEGFR-1 and VEGFR-2 in serum,BALF and lung tissue of rats was tested by using HE staining,immunohistochemitry and ELISA.Results:Compared with model group,the muscular artery morphology and lung pathology of rats in treatment group were improved,and the expression of VEGF,VEGFR-1 and VEGFR-2 in serum,BALF and lung tissue of rats in treatment group was decreased(P〈0.05).Conclusion:Baofei Dingchuan Decoction could improve the airway remodeling of COPD rats,and the mechanism might relate with the inhibition effect on the expression of VEGF,VEGFR-1and VEGFR-2 of VEGF signal pathway and the angiogenesis.
