摘 要:目的:探讨无花果叶提取物对OPG小鼠的抗骨质疏松作用及机制。方法:8周龄OPG基因敲除雌性小鼠60只,随机分成:实验组(无花果叶提取物组)、对照组(福善美组)、空白组(0.9%氯化钠溶液组),分别于喂药30、60d时用HE染色方法观察各组间骨小梁变化;用免疫组化法研究骨组织中白介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)的定位及信号表达强弱。结果:灌胃60d后实验组、对照组与空白组比较,骨小梁数目增加,差异有统计学意义(P<0.05);灌胃60d后实验组、对照组与空白组比较,IL-1和TNF-α阳性表达的细胞数目减少(P<0.05)。结论:无花果叶提取物具有抗骨质疏松作用,此作用可能与降低OPG基因敲除小鼠骨组织中IL-1、TNF-α的阳性细胞表达有关。Objective:To investigate the anti-osteoporosis effect and mechanism of fig leaf extraction on OPG knock out mice.Methods:Sixty eight-week old female OPG mice were randomly divided into three groups as fig leaf extraction group,fosamax group and control group.HE staining method was used to observe the changes in bone trabecula of mice in each group,and immunohis to chemistry method was used to observe the location and expression of IL-1 and TNF-α in bone tissue after treating for 30 d and 60 d.Results:The difference in the number of bone trabecula among fig leaf extraction group,fosamax group and control group was significant(P〈0.05) after treating for 60 d.The number of IL-1 and TNF-α positive cells in mice of fig leaf extraction and fosamax groups was less than that of control group(P〈0.05).Conclusion:Fig leaf extraction has the effect of antiosteoporosis,which might relate with the lower expression of IL-1 and TNF-α in OPG mice.
