摘  要:目的:探讨中药茵陈蒿的主要成分蒿属香豆素对大鼠胰腺星状细胞(PSCs)氧化、活化及细胞外基质沉积的影响。方法:分离PSCs并培养,免疫荧光法鉴定细胞,培养并传代至第3-5代后进行实验,分成空白组、丹酚酸B组、不同浓度香豆素组。分别培养24、48h后MTT法检测PSCs增殖情况。48h后通过比色法检测各组细胞丙二醛(MDA)、超氧化物歧化酶(SOD)水平的变化。免疫荧光、Western blot法检测各组平滑肌肌动蛋白(α-Smooth muscle actin,α-SMA)、I型胶原表达的情况。结果:与空白组相比,蒿属香豆素组的细胞增殖水平、MDA水平、α-SMA、Ⅰ型胶原的表达明显降低,SOD水平显著增高(P<0.05)。结论:蒿属香豆素能有效降低PSCs的氧化应激水平,抑制其活化进而减轻纤维化。Objective:To investigate the influence of scoparone on oxidation,activation and fibrosis of pancreatic stellate cells and deposition of extracellular matrix.Methods:PSCs were separated and cultivated.PSCs were identified by immunofluorescence method.The cells could be experimented when cultured in 3-5 generations.PSCs were divided into blank control group,positive control group and different concentrations of scoparone groups.After 24 h and 48 h,the cells proliferation was measured by MTT assay.The levels of intracellular malondialdehyde(MDA) and superoxide dismutase(SOD) were detected by colorimetric method after 48 h.The expression of α-Smooth muscle actin(α-SMA) and Collagen α were identified by immunofluorescence and Western blot.Results:Compared with blank control group,the levels of cells proliferation and intracellular MDA,and also the expression of α-SMA and Collagen:in the experimental groups decreased significantly,while the levels of SOD increased(P〈0.05).Conclusion:Scoparone could reduce the level of oxidative stress,suppress the activation of PSCs and alleviate fibrosis.
