摘 要:目的通过对Peroxiredoxin-1 (PRDX1), Glutathione S-transferase P1 (GSTP1)、和Glutathione Peroxidase-1 (GPX1)基因及其相关蛋白在儿童先天性肾积水肾组织中的表达水平的研究,探讨PRDX1、GSTP1和GPX1与氧化应激及梗阻性肾病发生、发展的关系。方法应用荧光实时定量PCR(qRT-PCR)、蛋白质印迹(Westernblot)和免疫荧光方法检测PRDX1、GSTP1和GPX1在15例肾积水患儿肾组织及15例肾胚瘤患儿瘤旁正常肾组织中的表达情况。结果PRDX1在肾积水肾组织中mRNA的表达量是正常对照组的1.14倍;GSTP1在肾积水肾组织中mRNA的表达量是正常对照组的1.09倍;GPX1在肾积水肾组织中mRNA的表达量是正常对照组的1.51倍,差异有统计学意义(P〈0.05)。Westernblot结果显示,PRDX1在肾积水肾组织中蛋白的相对表达量为103.36±2.94,高于正常对照组中的蛋白相对表达量;GSTP1在肾积水肾组织中蛋白的相对表达量为39.42±1.15,高于正常对照组中的蛋白相对表达量;GPXl在肾积水肾组织中蛋白的相对表达量为43.86±3.83,高于正常对照组中的蛋白相对表达量,差异有统计学意义(P〈0.05)。免疫荧光结果显示,PRDX1、GSTP1和GPX1在肾积水肾组织和正常对照组的肾组织中的肾小管及肾小球细胞胞质内呈阳性表达,三种蛋白的荧光强度在肾积水的肾组织中的表达均明显强于正常对照组。结论PRDX1、GSTP1和GPXlmRNA和蛋白在先天性肾积水的肾组织中表达上调,它们调节细胞的氧化应激水平,可能与氧化应激及梗阻性肾病的发生、发展有密切关系。Objective To explore the causes of hydronephrosis by studying the expression of peroxiredoxin-1 ( PRDX1 ), glutathione S-transferase P1 ( GSTP1 ) and glutathione peroxidase-1 (GPXl) genes and their proteins in renal tissues of hydronephrosis and Wilm's tumor. Methods The expressions of PRDX1, GSTP1 and GPX1 genes were detected by quantitative real-time polymerase chain reaction (qRT-PCR). Western blot and immunofluorescence were also used to detect the differential expressions of PRDX1, GSTP1 and GPX1 proteins between hydronephrosis and control groups. Results The expressions of PRDX1, GSTP1 and GPX1 genes were 1.14, 1.09 and 1.51-fold in hydronephrosis and control groups respectively. Similarly, the higher expression of PRDX1, GSTP1 and GPX1 proteins (103. 36 ±2. 94, 39. 42 ± 1.15 &43. 86 ± 3.83 respectively) in hydronephrosis were detected by Western blot. And immunofluorescence showed that PRDX1, GSTP1 and GPX1 had a significant increase in cytoplasm of tubules and glomeruli from hydronephrosis versus control group. Conclusions Higher expression of PRDX1, GSTP1 and GPX1 genes in hydronephrosis suggests that regulations of oxidative stress are probably involved in the occurrence and pathogenesis of obstructive nephropathy.
