摘  要:采用荧光光谱法、微分脉冲伏安法和紫外可见光谱法相结合研究了那格列奈与牛血清白蛋白(BSA)的相互作用机制。考察了不同环境温度和酸度那格列奈与牛血清白蛋白的结合常数和结合位点,并对其热力学常数进行了研究。结果表明,两者结合距离为4.29 nm,那格列奈主要通过疏水作用和静电作用与牛血清白蛋白相作用,造成牛血清白蛋白二级结构的变化。静态猝灭和非辐射能量转移是导致牛血清白蛋白荧光猝灭的原因。此外,乙醇会对二者的结合作用产生影响。为那格列奈在生物体内的储存、传输及药理作用提供一些重要的信息。The interaction between nateglinide( NG) and bovine serum albumin( BSA) was studied by fluorescence spectrometry, differential pulse voltammetry and UV-visible spectrophotometry. The binding constants,binding sites and thermodynamic parameters in different temperatures and acidity were studied. The results showed that the binding distance was 4. 29 nm. The hydrophobic force and electrostatic force played the main role in combination,which resulted in secondary structure changes of BSA. The fluorescence quenching of BSA was resulted from non-radiative energy transfer and static quenching. In addition,the effect of ethanol on binding with NG and BSA was investigated. The results are helpful to understand the storage,transmission and pharmacological effects on binding of NG with BSA.
