摘 要:目的:观察艾炷灸神阙穴对衰老豚鼠肝组织超氧化物歧化酶(Superoxide Dismutase,SOD)蛋白表达及基因表达的影响。方法:将健康雄性实验豚鼠随机分为正常组、模型对照组和治疗组,采用D-半乳糖腹腔注射建立豚鼠亚急性衰老模型,对治疗组进行艾柱灸治疗,每天1次,每次灸3壮,连续28d。用实时荧光绝对定量聚合酶链反应(Realtime Polymerase Chain Reaction,RT-PCR)法检测各组肝组织SOD相关基因表达,用免疫组化染色观察各组SOD蛋白表达。结果:造模后除正常组外,其余各组豚鼠血清SOD含量均较造模前明显降低(P<0.05),提示造模成功。治疗后治疗组SOD基因表达量明显高于模型组(P<0.05),与正常组差异无统计学意义(P>0.05)。肝组织免疫组化染色示:治疗组切片阳性率明显高于模型组(P<0.05),与正常组差异无统计学意义(P>0.05)。结论:艾炷灸神阙穴可提高衰老豚鼠肝组织SOD蛋白表达及相关基因表达。Objective: To observe the effects of moxa cone moxibustion at Shenque point on the protein and gene expression of superoxide dismutase( SOD) in the liver tissue of aging guinea pigs. Methods: Healthy male guinea pigs were randomly divided into normal group,model group,and treatment group. The subacute aging guinea pig model was established by intraperitoneal injection of D-galactose. In the treatment group,three moxa cones were used for Shenque point once a day for 28 consecutive days. The gene and protein expression of SOD in liver tissue was determined in each group using real-time PCR and immunohistochemical staining,respectively. Results: After model establishment,the serum content of SOD was significantly reduced in all groups( P〈0. 05) except the normal group,suggesting that the model was successfully established. After treatment,the treatment group had significantly higher SOD gene expression than the model group( P〈0. 05),while there was no significant difference in SOD gene expression between the treatment group and the normal group( P〉0. 05). Immunohistochemical staining of liver tissue demonstrated that the treatment group had a significantly higher positive rate of slices than the model group( P〈0. 05),but there was no significant difference in positive rate of slices between the treatment group and the normal group( P〉0. 05). Conclusion: Moxa cone moxibustion at Shenque point can elevate protein and gene expression of SOD in liver tissue of aging guinea pigs.
