半夏厚朴汤对慢性束缚应激小鼠行为学和神经递质含量的影响

摘　　要：目的:探讨半夏厚朴汤(BXHPT)对慢性束缚应激小鼠行为学和脑内神经递质含量的影响。方法:将60只小鼠随机分为正常对照组,模型组,半夏厚朴汤高、中、低剂量组,每组12只。用特制束缚桶束缚小鼠17d,每天6h,制备慢性束缚应激小鼠模型;每天造模前1h进行灌胃,半夏厚朴汤高、中、低剂量组分别灌胃半夏厚朴汤药液3.12、1.56、0.78g/m L,剂量为0.2m L/20g,空白组、模型组给予等量0.9%氯化钠注射液灌胃,连续给药18d。第18天灌胃结束后采用旷场实验和悬尾实验测定小鼠行为学改变,断头取出全脑,采用酶联免疫法测定小鼠不同脑区神经递质含量。结果:与对照组相比,模型组行为学指标(包括6min内不动时间、直立次数、修饰次数、穿格次数)及神经递质(5-HT、NE)含量均存在显著变化,差异具有统计学意义(P<0.01),表明模型建立成功。高剂量组各项指标与模型组相比,差异无统计学意义(P>0.05),中剂量组6min内不动时间、穿格次数、NE含量与模型组相比差异有统计学意义(P<0.05),直立次数、修饰次数、5-HT含量与模型组差异无统计学意义(P>0.05)。低剂量组各项指标均较模型组有显著改善,差异有统计学意义(P<0.05)。结论:半夏厚朴汤具有改善慢性束缚引起的小鼠行为学异常及下调脑内单胺类神经递质含量变化的作用。Objective： To investigate the effect of Banxia Houpo Tang（ BXHPT） on the behavior and intracephalic neurotransmitter content in mice with chronic restraint stress. Methods： Sixty mice were randomly and equally divided into normal control group,model group,and high-,middle-,and low-dose BXHPT groups. A mouse model of chronic restraint stress was prepared by restricting each mouse with a tailor-made restraint barrel 6 hours per day for 17 days. Intragastric administration was performed 1 h before model establishment every day; the high-,middle-,and low-dose BXHPT groups were given 3. 12,1. 56,and 0. 78 g / ml BXHPT（ 0. 2 ml /20 g）,respectively,by gavage,and the control group and model group received an equal volume of 0. 9% sodium chloride injection by gavage. All mice received intragastric administration for 18 days. After gavage on the 18 th day,the open field test and tail suspension test were used to determine the behavioral changes of the mice and the mice were decapitated to remove the whole brain. Enzyme-linked immunosorbent assay was applied to measure the neurotransmitter levels in different brain regions of the mice. Results： Compared with the control group,the model group had significant changes in behavioral indices（ including immobility time within 6 minutes,rearing times,modification times,and times of spanning lattice） and neurotransmitter（ 5-hydroxytryptamine（ 5-HT） and norepinephrine（ NE）） levels（ P〈 0. 01）,which suggested that the model was successfully established. No significant differences were found in any index between the high-dose BXHPT group and the model group（ P〉0. 05）. Between the middle-dose BXHPT group and the model group,immobility time within 6 minutes,times of spanning lattice,and NE level varied significantly（ P〈0. 05）,but there were no significant differences in rearing times,modification times,and 5-HT level（ P〉0. 05）. Compared with the model group,the low-dose BXHPT group showed significant improvements in all the indices（
