降脂理肝汤对非酒精性脂肪肝大鼠脂联素、SOD、TNF-α的影响

摘　　要：目的:观察降脂理肝汤对非酒精性脂肪肝大鼠脂联素、超氧化物歧化酶(SOD)、肿瘤坏死因子(TNF-α)的影响。方法:将61只SD雄性大鼠分为正常组(13只)和造模组(48只),经高脂饮食诱导造模,经切片成功后,将剩余40只造模组大鼠随机分成模型组,降脂理肝汤高、中、低剂量组(9.2、4.6、2.3g生药/kg),16周后取材,检测脂联素、SOD、TNF-α含量。结果:与模型组比较,降脂理肝汤各组均可明显增加脂联素的含量(P<0.01),中、高剂量组可明显提高SOD活性(P<0.01),高剂量组可明显降低TNF-α含量(P<0.01);与正常组比较,模型组与降脂理肝汤低、中剂量组脂联素含量均明显减少(P<0.01)、SOD活性明显降低(P<0.01),模型组TNF-α含量明显升高(P<0.01)。结论:降脂理肝汤可通过增加脂联素含量,增强胰岛素敏感性,提高SOD活性,降低血清TNF-α含量,抗氧化应激和减轻炎症治疗非酒精性脂肪肝,且其剂量与脂联素、SOD有明显相关性。Objective： To observe the effects of Jiangzhi Ligan decoction on adiponectin,superoxide dismutase（ SOD）,and tumor necrosis factor（ TNF）-α in rats with non-alcoholic fatty liver disease. Methods： Sixty-one male Sprague-Dawley rats were divided into normal group（ 13 rats） and modeling group（ 48 rats）. The model was induced by high-fat diet,and after sectioning,the remaining 40 rats in the modeling group were randomly divided into model group and high-,middle-,and low-dose Jiangzhi Ligan decoction groups（ treated with the crude drug at 9. 2,4. 6,and 2. 3 g / kg,respectively）. Sampling was made 16 weeks later to determine the levels of adiponectin,SOD,and TNF-α. Results： Compared with the model group,each Jiangzhi Ligan decoction group had a significantly increased adiponectin level（ P〈0. 01）,the middle-and high-dose Jiangzhi Ligan decoction group had a significantly increased SOD activity（ P〈0. 01）,and the high-dose Jiangzhi Ligan decoction group had a significantly reduced TNF-α level（ P〈0. 01）. Compared with the normal group,the model group and the low-and middle-dose Jiangzhi Ligan decoction groups had significantly reduced adiponectin level and SOD activity（ P〈0. 01）,and the model group had a significantly increased TNF-α level（ P〈0. 01）. Conclusion： Jiangzhi Ligan decoction can increase the sensitivity of insulin by raising adiponectin content and resist oxidative stress and alleviate inflammation by increasing SOD activity and reducing serum TNF-α level to treat non-alcoholic fatty liver disease. And the dose of Jiangzhi Ligan decoction is significantly correlated with adiponectin and SOD.
