摘 要:目的了解吉林省2013年柯萨奇病毒A组16型(CVA16)流行特征、种系进化及基因分型。方法对吉林省2013年分离获得的9株CVA16阳性毒株的VP1编码区核苷酸进行扩增及序列测定,使用Mega 4.0和Bioedit软件进行序列分析。结果 9株CVA16流行株中,1株为B1a基因亚型,与3株B1a代表株相比,其核苷酸和氨基酸的同源性分别为95.6%~96.8%和99.6%;8株为B1b基因亚型,与3株B1b代表株相比较,其核苷酸和氨基酸的同源性分别为94.1%~96.1%和98.3%~100%。2013年流行株与代表株相比共发生了9处氨基酸位点变异,其中8株B1b亚型中有6株在第23位氨基酸位点发生了亮氨酸→蛋氨酸的变异。结论 B1b是引起吉林省2013年CVA16流行的主要基因亚型,并且有大部分CVA16流行株在第23氨基酸位点发生了共同变异。Objective To understand the coxsackievirus group A type 16( CVA16) epidemic characteristics,phyletic evolution and genotyping in Jilin in 2013. Methods To amplify and analyze the sequence of the nucleotide in VP1 of 9 CVA16 positive strains isolated from Jilin in 2013,and used Mega 4. 0 and Bioedit software for sequence analysis. Results Out of the 9CVA16 strains,1 strain was B1 b subtype,and compared with the other 3 B1 b representative strains,its homology of nucleotide and amino acid were 95. 6%- 96. 8% and 99. 6%; 8 strains were B1 b subtypes,and compared with the other 3 B1 b representative strains,their homology of nucleotide and amino acid were respectively 94. 1%- 96. 1% and 98. 3%- 100%. Of the epidemic strains,compared with the representative strain,mutation occured on 9 amino acid sites,and out of 8 B1 b subtypes,the variation of Leucine→Methionine happened on the 23 amino acid site in 6 strains. Conclusion B1 b is the major gene subtype of CVA16 in Jilin in 2013,and there is a common variation on the 23 amino acid site of most CVA16 epidemic strains.
