摘 要:荧光定量PCR作为一种核酸定量技术,可在PCR扩增时在体系中加入荧光染料或荧光基团,实时监测荧光信号从而对PCR扩增产物进行实时准确定量,技术灵敏度高,特异性强。通过对荧光定量PCR的原理、分类、优缺点及几种定量分析方法对比进行简述,并对其在国内外海洋环境污染监测中的广泛应用和研究进行了全面综述,从而对荧光定量PCR和分子标志物在海洋监测中的应用前景做进一步的展望。Real-time fluorescent quantitative PCR(q RT-PCR) which works as one kind of quantitative techniques about nucleic acid with high sensitivity and strong specificity, refers to joining the fluorescent residues or fluorescence dye into PCR reaction system, and to real-time monitor entire PCR process using the fluorescence signal accumulation, and finally to real-time monitor the PCR amplified products. In this paper, the principle, classification, characteristics and the comparison of several analytical methods of q RT-PCR were reviewed; at the same time the wide application and research in marine pollution monitoring at home and abroad were also reviewed. In addition, they also predicted the further application of q RT-PCR and molecular markers in marine pollution monitoring.
