摘　　要：为了研究分枝杆菌噬菌体裂解酶lys A的生物学特性及对宿主菌的作用,对分枝杆菌肌尾噬菌体CJAUS5的lys A基因进行PCR扩增、克隆和原核表达,并利用生物信息软件对其序列进行分析。结果显示,CJAUS5-lys A基因与Gen Bank中分枝杆菌肌尾噬菌体的lys A基因具有高度同源性;克隆基因在大肠杆菌BL21中获得了成功表达,重组蛋白以包涵体的形式存在,蛋白分子量约为52 k Da;lys A蛋白保守区域分析结果显示,lys A包含具有酰胺酶活性的肽聚糖识别蛋白(PGRP)超家族,表明Lys A可能具有裂解肽聚糖的能力。本研究克隆表达出分枝杆菌噬菌体CJAUS5裂解酶Lys A,为进一步研究其相关功能奠定了基础。To investigate the biological characteristics of mycobacteriophage lys A protein and its effect on the host,a fragment of lys A gene was cloned and subsequently expressed in E.coli BL21.The sequence was analyzed by the bioinformatics software.The results showed that lys A gene shared high sequence similarity with other myoviridae mycobacteriophages.The lys A protein was successfully expressed in E.coli BL21 with the form of inclusion body.Meanwhile,the lys A protein contained the conservative region of PGRP superfamily which may have the activity of amidase.
