摘 要:目的:探讨温肾健脾方对糖尿病肾病(DN)大鼠肾小球足细胞的保护机制,为温肾健脾方在临床应用提供实验依据。方法:将链脲佐菌素诱导的DN大鼠30只,随机分为5组,即模型对照组、低剂量组(温肾健脾方7.5 g/kg)、中剂量组(温肾健脾方15 g/kg)、高剂量组(温肾健脾方30 g/kg)和阳性对照组(缬沙坦25 mg/kg);每组6只,每笼3只饲养,各组大鼠每天给予相应的药物一次,另外6只大鼠作正常对照组,每笼3只饲养。连续给药8周后,用透射电镜观察肾小球足细胞的结构。结果:与正常对照组比较,模型对照组大鼠肾小球足细胞足突消融,结构模糊;低、中、高剂量组大鼠足突均较完整、结构清晰、排列整齐,阳性对照组足突消抹、结构模糊。结论:温肾健脾方能保护肾小球和足细胞结构的完整性,可能是通过对DN大鼠肾小球结构产生作用从而发挥肾脏的保护,以控制蛋白尿并保护肾脏。Objective: To investigate the protective mechanism of warming kidney and invigorating spleen Recipe on DN ratglomerular podocytes. In order to provide experimental basis for its clinical application in. Methods: DN rats were induced by streptozotocin in 30,were randomly divided into 5 groups. That is the model control group,low- dose group( warming kidney and invigorating spleen Recipe 7. 5 g / kg) 、dose group( warming kidney and invigorating spleen Recipe 15 g / kg) 、high- dose group( warming kidney and invigorating spleen Recipe 30 g / kg) and positive control group( Valsartan 25 mg / kg); 6 rats in each group,each 3 cage rearing,each rat was given corresponding drugs once a day,the other 6 rats as normal control group and each 3 cage rearing. After 8weeks treatment,the structure of glomerular podocytes observed by transmission electron microscope. Results: Compared with the normal control group,model control group glomerular podocyte foot processes ablation and the structure of fuzzy; Low,medium and high dose group,all rats foot process is complete、structure clear and neat rows. Positive control group erasing foot processes,fuzzy structure. Conclusion: Warming kidney and invigorating spleen can protect the integrity of the glomerular and podocyte structure,it may be generated through the effect on glomerular structure in DN rats so as to exert renal protection which is in order to control the proteinuria and protect renal.
