摘 要:目的:首次探索建立含苦参中成药(当归苦参丸和四味土木香散)中黄酮类成分的定性鉴别方法。方法:采用薄层GF254硅胶板,鉴别三叶豆紫檀苷成分:以石油醚-乙酸乙酯-甲醇(5∶7∶1)溶液为展开剂,喷以10%的硫酸乙醇溶液,挥干,再喷以10%的磷钼酸溶液,加热至斑点显色清晰,置日光下检视各样品与三叶豆紫檀苷标准品比较结果;鉴别苦参酮成分:以石油醚-乙酸乙酯-甲醇(5∶5∶1)溶液为展开剂,挥干,置紫外光(365nm)下检视各样品与苦参酮标准品比较结果。结果:建立苦参中黄酮类成分三叶豆紫檀苷和苦参酮的薄层鉴别方法,在与对照品斑点相对应的位置上,供试品色谱显示相同颜色的斑点,且阴性无干扰。结论:该研究所建立的方法重复性高,操作简便,可用于薄层鉴别当归苦参丸和四味土木香散,可为其他含苦参中成药的定性鉴别方法提供参考。Objective:To establish thin layer distinction method for flavonoid compounds in Traditional Chinese Medicine(TCM)patent prescriptions containing with Sophora Flavescens flavonoid compounds.Methods:Using the silica GF254 thin layer plate.Trifolirhizin identification:used petroleum-ether-ethyl acetate-methanol(5∶7∶1)as the developing solvent,sprayed with 10%sulfuric acid in ethanol,evaporated to dryness,and then sprayed with 10% phosphomolybdic acid solution,heated the silica plate to spot the color clear,viewed the results of the sample comparing with trifolirhizin under the sunlight;Kurarinone identification:used petroleum-ether-ethyl acetate-methanol(5∶5∶1)as the developing solvent,evaporated to dryness,view the results of the sample comparing with Kurarinone standard under the UV(365nm)lamp.Results:Established a TLC identification method for Trifolirhizin and Kurarinone of Sophora Flavescens,in the chromatogram,comparing with the reference spot corresponding position was the same color of the spots,and negative non-interference.Conclusion:The method was repeatable and simple,could be used for the TLC identification of Danggui Kushen pills and Siwei Tumuxiang powder,and also provided a reference for the other TCM patent prescriptions containing with Sophora flavescens flavonoid compounds.
