摘 要:以阻止熟蚕吐丝的方法建立家蚕高蛋白血症模型,研究模型家蚕丝腺退化过程中细胞程序性死亡的特点,探索家蚕高蛋白血症模型的发病机制。形态解剖学观察显示,模型家蚕的丝腺退化过程延长了2倍时间;组织切片HE染色显示,模型家蚕的中部丝腺和后部丝腺都出现了退化启动推迟的现象。对家蚕组织解体退化信号通路相关基因表达水平的检测显示,模型家蚕丝腺细胞中自噬信号基因Atg6和Atg8的转录水平显著低于对照组家蚕,而凋亡信号基因Dronc的转录水平则显著高于对照组家蚕。免疫组化染色显示,模型家蚕丝腺细胞中凋亡启动蛋白Caspase-3在丝腺退化过程中持续表达。研究结果表明高蛋白血症模型家蚕丝腺细胞的凋亡活动增强,而丝腺退化延缓与自噬作用启动延迟有关。To investigate the pathological mechanism of hyperproteinemia,an animal model of hyperproteinemia has been created by blocking spinning process of the mature silkworm( Bombyx mori) larvae,upon which the characteristics of programmed cell death have been studied during the degradation process of silk gland( SGDP) in hyperproteinemia silkworm. The results from anatomical conformation and HE staining showed that the SGDP of hyperproteinemia silkworm was 2 times to that of the control ones,and the degradation process was delayed in middle silk glands and posterior silk glands. The detection results of expression level of related genes involved in degradation and decline signal pathway of silkworm tissues demonstrated that the transcription level of autophagy-related genes Atg6 and Atg8 in silk gland cells of hyperproteinemia silkworm were significantly lower than those of control,while the transcription level of apoptosis-related gene Dronc was significantly higher than that of control. Moreover,immunohistochemical staining indicated that Caspase-3 protein,a key apoptosis initiation related-protein,was expressed continuously at high level in the SGDP of hyperproteinemia silkworm. As a consequence,the apoptosis of silk gland cells in hyperproteinemia silkworm had been enhanced,while the retard of silk gland decline was related to start-up delay of autophagy.
