摘  要:目的:研究齐墩果酸(OA)对过氧化氢(H2O2)诱导的体外培养的人气道平滑肌细胞(ASMC)氧化应激损伤的保护作用。方法:以体外培养的ASMC为标本,以不同浓度的OA干预,经H2O2诱导损伤后,采用LIVE/DEAD染色、MTT实验、流式细胞仪、超氧化物歧化酶(SOD)测试盒分别检测实验及对照组细胞活性、细胞增殖、活性氧(ROS)的表达和SOD的活性。结果:加入20μmol/L的OA后,细胞的生长受到抑制;随着OA干预浓度的增加,死亡细胞逐渐减少;经OA预处理后,不同OA浓度组的ROS表达明显减低,SOD活性明显增加。结论:一定浓度的OA干预ASMC后,能抑制H2O2诱导的细胞增殖,对其诱导的氧化应激损伤有保护作用,OA可能具有干预哮喘气道炎症及气道重塑的作用。AIM: To evaluate the function of oleanolic acid( OA) protecting the oxidative stressmediated injury induced by hydrogen peroxide( H2O2) on human airway smooth muscle cell( ASMC) in vitro. METHODS: Human airway smooth muscle cells cultured by tissue adherence method from the trachea were treated with different concentrations of OA,then induced oxidative stressmediated injury by H2O2, using the method of LIVE / DEAD staining,MTT test and flow cytometry and superoxide dismutase( SOD) assay kit,detecting the cell activity,cell proliferation,reactive oxygen species( ROS) expression and the activity of SOD between the experiment and control groups.RESULTS: Cell proliferation was restrained after pretreating by 20 μmol / L OA. With the increase of concentration of OA,dead cells gradually reduced.After OA pretreatment,ROS expression was obviously reduced and SOD activity increased significantly in different concentrations of OA groups.CONCLUSION: A certain concentration of OA can inhibit the cell proliferation induced by H2O2,and has a protective effect on the oxidative stress injury induced by H2O2. Thus,OA may have the effect on airway inflammation and remodeling of asthma.
