摘 要:目的:本文对糖尿病心肌病发病的细胞外基质降解与重构机制进行探讨,为相关新药研发与临床治疗提供借鉴。方法:对本院诊治的糖尿病心肌病患者进行分析,检测糖尿病心肌病患者心功能及血清中丙二醛(MDA)和超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)水平变化,ELISA法检测细胞外基质降解与重构的酶类基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶组织抑制剂-1(TIMP-1)、基质金属蛋白酶组织抑制剂-2(TIMP-2)水平变化。以本院同期的健康体检者作为对照组。结果:与对照组相比,糖尿病心肌病患者心功能存在异常,且患者血清氧化应激分子水平明显升高,而抗氧化分子水平明显降低,同时基质金属蛋白酶表达明显升高,而基质金属蛋白酶抑制剂水平明显下降,与对照组比较,差异具有统计学意义(P<0.05)。结论:细胞外基质降解与重构参糖尿病心肌病的发病过程,且可能是药物治疗糖尿病心肌病的重要靶点。Objective:To explore the participation of extracellular matrix remodeling in the pathogenesis of diabetic cardiomyopathy.Methods:A total of 120 diabetic cardiomyopathy patients treated in our hospital were enrolled in this study.The serum concentration of MDA,SOD and GSHpx were analyzed in each group.The expression of MMP2,MMP9,TIMP1 and TIMP2were detected by ELISA.Results:The concentration of SOD and GSHpx was decreased greatly while the MDA was increased dramatically in diabetic cardiomyopathy patients when compared with control group.The results also showed that expression of MMP2 and MMP9were increased while the expression of TIMP1 and TIMP2content were decreased when compared with control.Conclusions:Extracellular matrix remodeling are involved in the pathogenesis of diabetic cardiomyopathy and they can be taken as targets for drug interventions.
