摘 要:目的:探讨γ-干扰素(IFN-γ)对哮喘小鼠气道炎症和肺组织中MUC5ac表达的影响。方法:40只雌性C57b L/6小鼠随机分为正常组、哮喘组、IFN-γ5万IU组和IFN-γ10万IU组,每组10只。测定各组小鼠呼吸道阻力,刘氏染色法分类计数支气管肺泡灌洗液(BALF)中细胞数,ELISA法检测BALF中IL-13的浓度,Ab-PAS染色计数气道上皮杯状细胞,PCR检测肺组织中MUC5ac mRNA表达。结果:与正常组相比较,哮喘组小鼠呼吸道阻力、BALF中细胞数,IL-13浓度明显增加,肺组织中MUC5ac表达升高(P<0.05);IFN-γ能减轻呼吸道阻力、BALF中细胞数,降低BALF中IL-13浓度和肺组织中MUC5ac表达(P<0.05)。结论 :IFN-γ能调节哮喘小鼠Th1/Th2平衡,抑制杯状细胞增生和肺组织中MUC5ac mRNA的表达。Objective To investigate the effects of IFN-γ on airway inflammation and protein MUC5ac mRNA expression in mice with asthma. Methods Forty c57bL/6 mice were randomly divided into control group, model group, IFN-γ 50,O00IU group and IFN-γ 100,000IU group, 10 for each group. The change rate of airway resistance was detected after acetylcholine chloride provocation. The count of inflammatory cells in BALF was analyzed. Levels of IL-13 in BALF were measured by ELISA. The lung tissue sections were stained with Ab-PAS for identification of goblet cells proliferation. Expressions of MUCSac mRNA in the lung tissues were determined by real time-PCR. Results As compared with the control group, the change rate of airway resistance, count of inflammatory cells, levels of IL-13 and MUCSac mRNA in lung were significantly increased in the model group (P 〈 0.05). IFN-γ reduced airway resistance, inflammatory cells, levels of IL-13 and MUCSac mRNA in the lungs (P 〈 0.05). Conclusions IFN-γ can regulate the balance of Thl/Th2, inhibit airway goblet cell proliferation, and protein MUC5ac mRNA expression in mice withasthma.
