摘 要:目的分析日本血吸虫感染以及虫源性糖蛋白果糖二磷酸醛缩酶(Sj FBPA)刺激后小鼠脾脏细胞中白介素4(IL-4)、白细胞分化抗原CD8a因子表达量的动态变化,并探讨Sj FBPA与辅助型T细胞(Th2)免疫应答的关系。方法血吸虫尾蚴感染C57小鼠10只,设正常对照组。感染8周后处死小鼠,取脾脏制备淋巴细胞悬液;用藻红蛋白(PE)标记CD8a抗体,藻青蛋白(APC)标记IL-4抗体进行细胞染色;流式细胞仪检测细胞因子表达量的变化。10只正常C57小鼠处死后取脾脏制成淋巴细胞悬液,加Sj FBPA蛋白体外刺激,按上述流程刺激方法检测细胞。结果小鼠感染血吸虫8周后脾细胞中IL-4、CD8a的表达量分别为(22.39±5.93)%、(11.34±2.15)%,正常对照组脾细胞中IL-4、CD8a的表达量分别为(1.39±0.52)%、(1.98±0.84)%,差异有统计学意义(P<0.01)。Sj FBPA体外刺激小鼠脾细胞的IL-4、CD8a的表达量分别为(14.14±1.16)%、(8.99±0.81)%,正常对照组脾细胞中IL-4、CD8a的表达量分别为(1.86±0.93)%、(1.69±0.56)%,差异有统计学意义(P<0.01)。结论 Sj FBPA在血吸虫感染中具有诱导Th2免疫偏移的作用,树突状细胞作为提呈细胞参与该免疫反应。Objective To analyze the dynamics of IL-4,CD8 a in spleen cells of mice infected with cercariae of Schistosoma japonicum or Sj FBPA and investigate the relationship between Sj FBPA and the Th2-dominant response.Methods 10 mice were infected by cercariae and 10 mice were uninfected. 8 weeks after infection,all mice were sacrificed. Spleen single cell suspension was prepared and analyzed by flowed cytometry. Analysis was performed on the IL-4,CD8 a cells of spleen cells stained with anti FITC-CD4,PE-CD8 a,APC-IL-4 monoclonal antibody. Results At 8 weeks,the percentage of IL-4 and CD8 a cells in infected with cercariae of Schistosoma japonicum mice was 24. 5% and 7. 65%,respectively,while the percentage of IL-4 and CD8 a cells in uninfected mice was 1. 18%and 0. 571%,indicating a gradual decline( P〈0. 05). The percentage of IL-4 and CD8 a cells in infected with Sj FBPA was 18. 3% and 5. 82%,the percentage of IL-4 and CD8 a cells in uninfected mice was 0. 91% and0. 086%,indicating a gradual decline( P〈0. 05). Conclusion This study suggests that Sj FBPA may have a potential value for the Th2 response. DC cells as presenting cells are involved in the immune response.
