摘 要:目的评价盐酸戊乙奎醚对新生大鼠内毒素性急性肺损伤时Toll样受体4(TLR4)mRNA表达的影响。方法清洁级健康雄性Wistar大鼠30只,体重12~18g,7日龄。采用随机数字表法分为3组(n=10):生理盐水组(NS组)、急性肺损伤组(ALI组)和盐酸戊乙奎醚组(PH组)。采用腹腔注射内毒素5mg/kg的方法制备大鼠内毒素性急性肺损伤模型,PH组腹腔注射盐酸戊乙奎醚0.5mg/kg后30min制备模型,NS组给予等容量生理盐水。于注射内毒素后dh时处死大鼠取肺组织,采用RT—PCR法检测TLR4mRNA表达,ELISA法测定TNF-α、IL-1β和IL-10的含量,光镜下观察肺组织病理学结果。结果与NS组比较,ALl组和PH组肺组织TLR4mRNA表达上调,TNF-α、IL-1β和IL-10含量升高(P〈0.05);与ALl组比较,PH组肺组织TLR4mRNA表达下调,TNF-α、IL-1β和IL-10含量降低(P〈0.05)。PH组肺组织病理学损伤较ALI组减轻。结论盐酸戊乙奎醚通过下调肺组织TLR4mRNA表达,抑制炎性反应,从而减轻新生大鼠内毒索性急性肺损伤。Objective To evaluate the effects of penehychdine hydrochloride on Toll-like receptor 4 (TLR4) mRNA expression during endotoxin-induced acute lung injury (ALI) in neonatal rats. Methods Thirty pathogen-free male Wistar rats, aged 7 days, weighing 12- 18 g, were randomly divided into 3 groups (n= 10 each) using a random number table: normal saline group (group NS), ALl group, and penehyehdine hydroehloride group (group PH). ALl was induced with intraperitoneal endotoxin 5 mg/kg. In group PH, penehychdine hydrochloride 0.5 mg/kg was injected intraperitoneally at 30 rain before ALl, while the equal volume of normal saline was given instead in group NS. At 4 h after endotoxin injection, the rats were sacrificed, and lungs were removed for determination of TLR4 mRNA expression (using real-time reverse transcriptase polymerase chain reaction) , and contents of tumor necrosis factor-alpha (TNF-α) , interleukin-1 beta (IL-1β) , and IL-10 in the lung tissues (by enzyme-linked immunosorbent assay) , and for examination of the pathologic changes of lung (with electron microscope). Results Compared with group NS, TLR4 mRNA expression was significantly up-regulated, and contents of TNF-α, IL-1β, and IL-10 were increased in ALI and PH groups. Compared with group ALI, TLR4 mRNA expression was significantly down-regulated, and contents of TNF-α, IL-1β, and IL-10 were decreased in group PH. The pathologic changes of lung were significantly mitigated in group PH compared with group ALI. Conclusion Penehychdine hydrochloride inhibits inflammatory responses through down-regulating TRL4 mRNA, thus reducing endotoxin-induced ALI in neonatal rats.
