补肾壮元胶囊对肾阳虚小鼠性激素水平及免疫功能影响的实验研究

摘　　要：目的:观察补肾壮元胶囊对肾阳虚模型小鼠性激素水平的影响及其对免疫功能的调节作用,探讨其临床作用机制。方法:将60只昆明种小鼠按照体质量随机分为6组:正常对照组、模型组、右归丸组、补肾壮元胶囊高、中、低剂量组。除正常组外,其他组小鼠均用氢化可的松注射液制备肾阳虚模型,正常组和模型组给予等体积的蒸馏水,各治疗组给予相应剂量的药物,持续给药20天。观察小鼠体质量和免疫器官及睾丸质量和性激素水平的变化,测定小鼠腹腔巨噬细胞吞噬功能、TNF-α分泌量及脾脏NK细胞活性。结果:模型组小鼠体质量、睾丸质量、性激素水平及小鼠胸腺指数、脾脏指数、腹腔巨噬细胞吞噬功能和NK细胞活性等指标较正常对照组显著降低(P<0.01);补肾壮元胶囊高、中、低剂量灌胃给药均可显著提高雄性小鼠体质量及免疫器官胸腺及脾脏质量(P<0.05或P<0.01),提高血清睾酮(T)、促黄体生成激素(LH)、促卵泡生成激素(FSH)水平(P<0.05或P<0.01),并增强巨噬细胞吞噬功能及其分泌TNF-α作用,增强NK细胞的活性(P<0.05或P<0.01);且大剂量优于小剂量。结论:补肾壮元胶囊具有补肾壮阳作用,能显著提高雄性小鼠性激素水平,对小鼠免疫功能有明显促进作用。Objective： To observe the effects of Bushen Zhuangyuan Capsule on sexual hormone level and immune function in kidney- yang deficiency mice and to investigate the mechanism of its clinical effect. Methods： 60 KM male mice were randomly divided into 6 groups. Except for normal group,all mice were injected hydrocortisone to establish the kidney- yang deficiency models. Mice in normal and model groups were fed with distilled water,and the others were given Bushen Zhuangyuan Capsule or Yougui Pill respectively for 20 days continuously. Then,the weight of testis,spleen and thymus were meseaused,and the blood levels of T,LH,FSH and the macrophage phagocytosis function,TNF- α secretion and the activities of NK cells were examined. Results： The weight of testis,spleen and thymus in model group were much lighter than those in normal,the blood level of T,LH,FSH,macrophage phagocytosis function and the activities of NK cells in model group were decreased to compare with normal control,（ P〈0. 01）. Bushen Zhuangyuan Capsule in high,medium and low doses could significantly improve the weight of testis,spleen and thymus of the male mice（ P〈0. 05 or P〈0. 01）,obviously increase the serum T,LH,FSH levels of the male mice（ P〈0. 05 or P〈0. 01）. It also obviously promoted the the macrophage phagocytosis function and the release of TNF- α and activity of NK cell（ P〈0. 05）; large dose worked better than small one. Conclusion： Bushen Zhuangyuan Capsule can benefiting kidney and enhancing yang,significantly improve sexual hormone level and immune function in mice.
