摘 要:目的研究富硒板桥党参(RSBCP)合剂对衰老小鼠免疫功能的影响。方法昆明种小鼠60只,雌雄各半,随机分为RSBCP合剂高剂量组、中剂量组、低剂量组、模型组及对照组,每组12只。对照组按500 mg/(kg·d)腹腔注射生理盐水,其他各组注射等量D-半乳糖。RSBCP合剂高、中、低剂量组在注射D-半乳糖的同时按(100、50、25)g/(kg·d)RSBCP合剂灌胃,模型组、对照组等量蒸馏水灌胃。30 d后,处死小鼠。测定胸腺指数、脾脏指数,ELISA检测各组小鼠血清中IgG、IgM、C3、C4、白细胞介素2(IL-2)、IL-6及肿瘤坏死因子α(TNF-α)含量,采用细胞流式术分析脾细胞主要T细胞亚群。结果与对照组相比,模型组胸腺指数、脾脏指数明显下降,IgG、IgM及C3、C4、IL-2、IL-6含量显著减少、TNF-α含量明显增加;与模型组比较,RSBCP合剂各组胸腺指数、脾脏指数显著增大,IgG、IgM及C3、C4、IL-2、IL-6含量显著增加、TNF-α含量减少;与对照组相比,RSBCP高剂量组胸腺指数、脾脏指数增大显著,IgG、IgM及C3、C4、IL-2、IL-6含量明显增加、TNF-α含量减少;中、低剂量RSBCP组胸腺指数、脾脏指数、IgG、IgM及C3、C4、TNF-α与对照组无显著性差异。与对照组相比,模型组CD3^+T细胞、CD4^+T细胞及CD44^+T细胞有明显减少,CD8^+T细胞明显增加;与模型组相比,RSBCP合剂各组CD3^+T细胞、CD4^+T细胞及高、中剂量RSBCP组CD44^+T细胞显著增加,CD8^+T细胞明显减少。结论 RSBCP合剂对衰老小鼠胸腺、脾脏萎缩有一定的延缓作用,能显著提高衰老小鼠血清中IgG、IgM、C3及C4含量,提升IL-2、IL-6水平,降低TNF-α水平,影响T细胞亚群的比例,对增强免疫功能有作用。Objective To investigate the effect of rich selenium-Banqiao-Codonopsis Pilosula (RSBCP) mixture on the immune functions of the aging mice. Methods Sixty Kunming mice (half male and half female) were randomly divided into high-, middle- and low-dose RSBCP mixture groups, model group and control group, with twelve mice in each group. The control group was injected intraperitoneally with 500 mg/( kg · d) normal saline and the other groups were injected with the same amount of D-galactose. The RSBCP mixture high-, middle- and low-dose groups were respectively gavaged with 100, 50, 25 g/(kg · d) RSBCP mixture at the moment when D-galactose was injected. The model group and control group were given the same amount of distilled water instead. The mice were killed at the 30th days and the indexes of thymus and spleen were measured. The serum levels of IgG, IgM, C3, CA, interleukine 2 ( IL-2), IL-6 and tumor necrosis factor alpha (TNF-α) were detected by ELISA and the major T-cell subsets of splenocytes were analyzed by flow cytometry. Results Compared with the control group, the indexes of thymus and spleen in the model group decreased obviously; the serum levels of IgG, IgM, (33, CA, IL-2 and IL-6 were reduced significantly and the level of TNF-α increased significantly. Compared with the model group, the indexes of thymus and spleen in the RSBCP mixture groups increased obviously; the levels of IgG, IgM, C3, CA, IL-2 and IL-6 were raised significantly and the level of TNF-α decreased significantly. Compared with the controlgroup, the indexes of thymus and spleen in the RSBCP mixture high-dose group increased obviously; the levels of IgG, IgM, C3, CA, IL-2 and IL-6 were lifted significantly and the level of TNF-α decreased significantly. The indexes of thymus and spleen and the levels of IgG, IgM, C3, CA, IL-2 and IL-6 in the RSBCP mixture middle- and low-dose groups had no obvious differences from those of the control group.
