桃红四物汤对光老化小鼠皮肤组织中MMP-1、MMP-3、MMP-12、Smad-2蛋白表达水平影响的实验研究

摘　　要：目的:研究桃红四物汤对光老化小鼠皮肤组织中MMP-1、MMP-3、MMP-12、Smad-2蛋白表达水平的影响。方法:将30只小鼠随机分到6个组中,分别为中药高、中、低剂量组、阳性对照组(VE组)、模型对照组、和空白对照组。模型对照组、VE组和中药低、中、高剂量组小鼠造皮肤光老化模型,空白对照组小鼠正常饲养。在造模前30 min,给各组小鼠灌胃,空白对照组和模型对照组给生理盐水,高、中、低剂量组给相应剂量药液,VE组给予维生素E滴剂。造模完成后,采用Western blot方法检测小鼠组织中MMP-1、MMP-3、MMP-12、Smad-2蛋白表达水平含量。结果:与空白对照组比较,模型对照组小鼠皮肤组织中MMP-1、MMP-3、MMP-12蛋白含量的表达水平显著上升,差异具有统计学意义(P<0.01),Smad-2蛋白表达水平显著下降(P<0.01);与模型对照组相比,中药低、中、高剂量组小鼠皮肤组织中MMP-1、MMP-3、MMP-12蛋白含量的表达水平显著下降,差异具有统计学意义(P<0.01),Smad-2蛋白表达水平显著上升,差异具有统计学意义(P<0.01)。结论:桃红四物汤对皮肤光老化具有一定的保护作用。Objective： To study the effect of Taohong Siwu Decoction on expressions of MMP- 1,MMP- 3,MMP- 12 and Smad- 2 protein in skin tissue of photoaging mice. Methods： Thirty mice were divided into 6 groups,high dose group,moderate dose group,low dose group,positive control group（ VE group）,model control group and blank control group. Blank control group was normally fed and the other groups were established photoaging model. Before 30 minutes of the preparation of photoaging model,we gave saline to blank control group and model control group,corresponding dose of drug liquid to high,medium and low dose groups,vitamin E drops to positive control group. The expression levels of MMP- 1,MMP- 3,MMP- 12 and Smad- 2 protein in skin tissue in each group were measured by Western- blot. Results： Compared with blank control group,expressions of MMP- 1,MMP- 3 and MMP- 12 protein in skin tissue of model control group were significantly higher（ P〈0. 01） and expressions of Smad- 2 protein in skin tissue of model control group were significantly lower（ P〈0. 01）. Compared with model control group,expressions of MMP- 1,MMP- 3 and MMP- 12 protein in skin tissue of high,medium and low dose groups were significantly lower（ P〈0. 01） and expressions of Smad- 2 protein in skin tissue of high,medium and low dose groups were significantly higher（ P〈0. 01）. Conclusion： Taohong Siwu Decoction has a protective effect on the skin photoaging.
