摘 要:目的:观察益气化瘀方对压迫损伤后不同时间点大鼠DRG神经元TRPV4基因表达的影响。方法:大鼠随机分为假手术组10只、模型组40只、中药组40只。模型组和中药组又根据取材、给药时间点的不同分为3 d组、7 d组、14 d组、28 d组,每组10只。用Von Frey法测试大鼠造模后50%撤足阈值;应用RT-PCR法检测大鼠DRG神经元TRPV4基因表达情况。结果:造模后,各模型组大鼠与假手术组相比50%撤足阈值均有显著性降低;中药干预后,在中药7d组、14 d组和28 d组大鼠50%撤足阈值均显著高于相应时间点各模型组。模型组各时间点TRPV4mRNA的表达与假手术组相比显著增高。中药干预后第3天、第7天和第14天组TRPV4mRNA的表达仍高于假手术组,而中药28 d组TRPV4mRNA的表达与假手术组相比无显著性差异。在第7天、第14天和第28天,中药干预组和模型组各时间点TRPV4mRNA表达相比均有显著性差异。结论:益气化瘀方可能通过下调TRPV4基因表达从而影响神经根压迫损伤后根性神经痛的产生。Objective: To observe the influence of Yiqi Huayu Formula on TRPV4 gene expression of dorsal root ganglion neurons of lumbar nerve root compression injury in rats. Methods: The rats were randomly divided into sham operation group( 10rats),model group( 40 rats) and TCM group( 40 rats). The model and TCM groups were divided into 3 d group,7 d group,14 d group,14 d group and 28 d group according to materials,administration time,10 in each. Von Frey method was used to detect the50% foot- withdrawing threshold and RT- PCR to detect the TRPV4 gene expression. Results: After modeling,compared with the sham operation group,the model groups' 50% foot- withdrawing threshold was significantly decreased. After TCM intervention,TCM 7 d,14 d and 28 d groups' 50% foot- withdrawing threshold was significantly higher than that of the model groups. Compared with the sham operation group,model groups' TRPV4 mRNA expressions at different time points were significantly increased. TCM 3 d,7 d and 14 d groups' TRPV4 mRNA expressions were higher than that of the sham operation group. TCM 28 d group's RPV4 mRNA expression had no difference compared with that of sham operation group. On 7,14 and 28 d,TCM and model groups' TRPV4 mRN expressions had significant difference. Conclusion: Yiqi Huayu Formula may down- regulate the TRPV4 gene expression to influence the nervous pain due to lumbar nerve root compression injury.
