组胺对人肝星状细胞合成Ⅰ型胶原的影响

摘　　要：研究组胺对人肝星状细胞(HSC-T6)合成Ⅰ型胶原的影响,探讨肥大细胞(MC)在慢性肝疾病中的作用机理。采用不同浓度(5×10-6、10×10-6、50×10-6、100×10-6mol/L)组胺刺激体外培养的HSC-T6;并设立不同时间点(24、36、48h)分别收集培养上清液,ELISA法测定培养上清液中的Ⅰ型胶原含量。结果表明,组胺能明显促进HSC合成分泌Ⅰ型胶原,且存在一定的时效关系和量效关系,MC可以通过其分泌的生物活性介质(组胺)增强HSC-T6分泌Ⅰ型胶原,而参与慢性肝病发生发展过程。The paper studied the effects of the histamine on collagen Ⅰ synthesis of human hepatic stellate cells line HSC-T6 cells. It explored the mechanism of mast cells（MC） involving in chronic liver diseases. HSC-T6 cells were co-cultured with the histamine of different concentrations（5 ×10-6, 10 ×10-6, 50 ×10-6, 100 ×10-6mol/L）in vitro. The supernatant was collected at different incubation period（24, 36, 48 h）. The content of collagen Ⅰin the supernatant was evaluated by the ELISA method. The histamine could enhance significantly collagen Ⅰprotein synthesis of HSC-T6. There were the tendency of time and comcentration. MC may participate in the progression of chronic liver diseases by the histamine, a kind of biologically active substance secreted by MC.
