摘 要:目的:探讨中药红景天及其主要成分红景天苷对高糖培养下血管平滑肌细胞(VSMCs)增殖的作用及机制。方法:将对数增殖期的VSMCs随机分为低糖(5 mmol/L)培养和高糖(25 mmol/L)培养两组,每组再分为5个亚组,分别在普通培养基(对照组)、低浓度红景天(0.3 mmol/L)、高浓度红景天(0.5 mmol/L)、低浓度红景天苷(0.3 mmol/L)、高浓度红景天苷(0.5 mmol/L)5种干预条件下培养24 h,采用BrdU法观察VSMCs增殖情况,Elisa法检测活性氧(ROS)水平和还原型烟酰胺腺嘌呤二核苷酸磷酸(NADPH)氧化酶活性。结果:低糖培养下各亚组间VSMCs增殖无差异,ROS水平和NADPH氧化酶活性亦无差异。在高糖培养条件下,VSMCs增殖明显,ROS水平和NADPH氧化酶活性明显升高;加入红景天/红景天苷后,VSMCs增殖、ROS水平和NADPH氧化酶活性明显被抑制,并且存在一定的浓度依赖性。结论:红景天及红景天苷可抑制高糖诱导的VSMCs增殖,可能与抑制氧化应激水平有关,红景天苷可能是红景天发挥这一作用的主要成分。Objective:To investigate the effects of rhodiola and its main composition salidroside on the proliferation of vascular smooth muscle cells(VSMCs)cultured in different levels of glucose and the possible mechanism. Methods:The VSMCs in logarithmic growth phase were randomized into two groups with low glucose(5 mmol/L)and high glucose(25 mmol/L)and there were five different interventions imposed on each group for 24 hincluding the normal medium(control group),the low concentration of rhodiola(0.3 mmol/L),high concentration of rhodiola(0.5 mmol/L),the low concentration of salidroside(0.3mmol/L)and high concentration of salidroside(0.5 mmol/L).BrdU method was used to determine the proliferation of VSMCs and Elisa assay was used to detect reactive oxygen species(ROS)levels and nicotinamide adenine dinucleotide phosphate(NADPH)oxidase activity. Results:In low glucose condition,there was no statistical significance in cell proliferation,ROS levels and NADPH activity among all subgroups.In high glucose condition,cell proliferation,ROS production and NADPH oxidase activity were more obvious than those in low glucose condition,and in rhodiola or salidroside subgroups,cell proliferation,ROS production and NADPH oxidase activity were apparently inhibited,and there was certain concentration dependence. Conclusion:Rhodiola or salidroside may inhibit high glucose-induced VSMCs proliferation possibly by inhibiting oxidative stress,and the effect of rhodiola may be mainly mediated by salidroside.
