Basic and translational study of extracellular microvesicles and exosomes in vascular diseases

摘  要:Exosomes (EXs, 30 - 120 nm) and microvesicles (MVs, 100 - 1000 nm) are two types of extracellular microparticles. Their contents (miRs and proteins) vary depending on cell origin and status. Mounting evidence suggest that EXs and MVs mark the characters of their parent cells and represent a novel mechanism of intercellular communication. Therefore, they can serve as potential biomarkers as well as therapeutic avenues. MiRs are small non-coding RNAs, regulating a variety of biological processes. Endothelial cells (ECs) dysfunction/injury is thought to be the common pathphysiology of cardiovascular diseases and the endothelial progenitor cells (EPCs) are reduced in number and dysfunctional in diabetic patients. Our group has reported that EPCs released EXs (EPC- EXs) can improve the angiogenic effects in injured ECs via delivering miR126. In the in vitro cell systems, we have demonstrated that cMVs from diabetic patients (d-cMVs) impaired EPC migration ability, and increased their apoptosis and ROS production, which are associated with the changes in the levels of miR-126, miR-155 and VEG- FR2 in cMVs and the h-EPCs after co-incubation. More recently, we have established a novel specific and sensitive method for isolation and NTA analysis of EXs based on magnetic activated cell sorting (MACS) and fluorescence (q-dots) NTA techniques. Further studies in this area will facilitate the discovery of novel biomarkers and thera- peutic avenues for various diseases.
