摘 要:目的:测定峨参提取物中的多糖含量并研究其抗氧化作用。方法:按照硫酸-蒽酮法,在分光光度计620 nm处测定吸光度,按标准曲线所得回归方程计算峨参多糖的含量;同时采用邻苯三酚在碱性条件下能发生自氧化的原理,根据抑制率=[A0-(Ax-Ai0)]/A0×100%,计算不同浓度的峨参多糖对超氧阴离子产生的抑制率来观察其抗氧化作用。结果:峨参提取物中多糖浓度为0.389 9 mg/m L,含量为38.99%;且在0.1-1 mg/m L浓度内,多糖对超氧阴离子的抑制率随多糖浓度的增加而增强,在0.6-1 mg/m L浓度之间,其抑制率明显增强,最大抑制率达到31.22%。结论:峨参提取物中的多糖含量较高,且具良好的体外抗氧化活性,为把峨参多糖作为天然抗氧化剂开发提供了科学依据。Objective: To study on determination and antioxidant activity of polysaccharides in EShen [Anthriscus sylvestris(L.) Hoffm] extracts. Methods: According to the anthrone-sulfuric acid method, the contents of polysaccharides in EShen extracts were calculated by regression equation obtained from the standard curve and the absorbance was measured under 620 nm with spectrophotometer; simultaneously, with the principle of pyrogallic acid happening auto-oxidation in alkaline situation, the inhibition rates of different concentrations of polysaccharides on superoxide anion were calculated to observe its antioxidant activity in the light of inhibition rate=[A0-(Ax-Ai0)]/AO ×100%. Results: The concentrations of polysaccharides in EShen extracts were 0.389 9mg/m L and 38.99%; the inhibition rate of polysaccharides in EShen extracts on superoxide anion increased as the concentrations of polysaccharides raised in the concentrations between 0.1 and 1 mg/m L, the inhibition rates enhanced obviously in the concentrations between 0.6 and 1 mg/ML, maximum inhibition rate reached 31.22%. Conclusion: Polysaccharides in EShen extracts are in high contents, which show better antioxidant activity in vitro, it would provide scientific basis for the development of polysaccharides in EShen extracts as a natural antioxidant.
