摘  要:目的观察转染微小RNA-21(mi R-21)对MCF7细胞系中磷酸酶和张力蛋白同源物基因(PTEN)表达及细胞凋亡的影响。方法对MCF7细胞转染mi R-21,采用免疫组织化学法检测转染前后PTEN的表达;用噻唑蓝(MTT)比色法测转染mi R-21后MCF7细胞增殖;流式细胞术检测转染后MCF7细胞凋亡。结果经免疫组织化学检测,转染组未见染色,对照组细胞胞质呈棕褐色,转染组PTEN的表达明显低于对照组,MTT法检测转染组细胞活力增加(P<0.05)。转染组的凋亡率为(3.14±0.24)%,空白组、空质粒组的凋亡率分别为(8.04±0.02)%、(9.41±0.53)%(P<0.05),转染组细胞凋亡率明显减少。结论 PTEN是mi R-21的靶基因之一,mi R-21的高表达可促进乳腺癌的发展。[ Objective ] To explore the expression of phosphatase tensin homologue (PTEN) and cell apoptosis in MCF7 cell line before and after microRNA-21 (miR-2) was transfected. [Methods] MiR-21 Was transfected in MCF7 cell line, Immunohistochemical detection of PTEN expression after transfection; transfected with miR-21 MCF7 cell proliferation measured by Tetrazolium (MTT) assay; flow cytometry detection of transfected MCF7 cells. [ Results ] The control group were detected by immunohistochemistry, cytoplasmic brown color. By transfection group showed no staining, transfected group PTEN expression was significantly lower than the control group. MTT assay transfected cell viability was significantly be promoted (P 〈 0.05). Transfection group apoptosis rate was (3.14 ± 0.24)%, and a blank group, the apoptosis rate of the empty plasmid group (8.04± 0.02) % (9.41 ± 0.53) % (P 〈 0.05), apoptosis of the transfected cells is significantly reduced. [ Conclusion] PTEN is a miR-21 target genes. The high expression of miR-21 can promote the development of human breast cancer.
