摘  要:采用分离纯化成功的胰岛细胞经PDGF因子处理后,MTT法、流式细胞术和胰岛素释放实验法检测猪胰岛细胞增值率,细胞周期及细胞功能的变化,探讨PDGF因子对滇南小耳猪胰岛细胞存活及功能的作用.结果显示,培养48 h后经PDGF因子处理的猪胰岛(实验组)细胞增值率、S期细胞的百分比、对高糖和低糖的刺激明显高于正常培养猪胰岛细胞(对照组)(P<0.05).因此,经PDGF因子处理后的滇南小耳猪胰岛细胞可能是通过加速猪胰岛细胞的DNA合成,促使细胞迅速进入DNA复制期,加速细胞分裂进而显著增加胰岛细胞增殖能力和功能.To objective the role of PDGF factor in the survival and their resistance of porcine islet cells. The isolated and purified islet cells were divided into 2 groups: normal cultured cells group( control group),and PDGF factor cultured group( experimental group). The proliferation rate in porcine islet cells was detected by MTT assay. The change of cell cycle and function were detected by flow cytometry and insulin release test. The Results that at 48 h,the rate of porcine islet cell proliferation,the percentage of cell in S-phase and the stimulation response to high sugar and low sugar were significantly higher in PDGF factor treated islet cells( experimental group) compared with normal cultured porcine islet cells( control group)( P〈 0. 05). So Diannan small ear pig islet cells treated PDGF factormay be accelerate cell division,and significantly increase the proliferation and function in islet cell,by increasing the DNA synthesis of porcine pancreatic islet cells,and promoting the rapid access to the DNA replication.
