益气补肾活血方对佐剂关节炎大鼠滑膜ERK1/2的影响

摘　　要：目的以益气补肾活血方治疗佐剂关节炎(adjuvant arthritis,AA)大鼠,通过观测该中药复方对AA大鼠滑膜(extracellular signal-regulated kinase 1/2,ERK1/2)表达水平的影响,探讨该中药复方对类风湿关节炎的治疗机制。方法以雷公藤多苷作为对照,采用AA大鼠模型,用益气补肾活血方对AA大鼠进行灌胃治疗。通过动态观测SD大鼠关节肿胀度及滑膜ERK1/2的磷酸化表达结果,并以灰度扫描软件分析结果,以观测该中药复方对AA大鼠滑膜ERK1/2表达的影响。结果益气补肾活血方既能明显降低AA大鼠的关节肿胀度,又能明显降低AA大鼠滑膜ERK1/2的磷酸化表达水平。结论滑膜ERK1/2的磷酸化表达水平在类风湿关节炎发病机制中具有重要作用,而益气补肾活血方能显著降低滑膜ERK1/2的磷酸化表达水平,可能是该方治疗类风湿关节炎的作用机制之一。Objective To explore the effects of Yiqi Bushen Huoxue（ YQBSHX） decoction on ex-tracellular signal-regulated kinase 1/2 （ ERK 1/2 ） expression in the synovial tissue in adjuvant arthritis （ AA） rats, and further to investigate the mechanism of Yiqi Bushen Huoxue decoction in the treatment of rheumatoid arthritis. Methods With the tripterygium glycosides as the control drug, the joint swelling degree of SD rats was observed dynamically, the level of phosphorylated expression of extracellular signal-regulated kinase （ ERK） 1/2 in the synovial tissue was detected by using the gray scale scanning software to analyze the effects of Yiqi Bushen Huoxue decoction. Results Yiqi Bushen Huoxue decoction can significantly reduce the joint swelling degree of arthritis rats, and the level of phosphorylated expression of ERK 1/2 in the synovial tissue. Conclusion The level of phosphorylated expression of ERK 1/2 in the synovial tissue played an important role in the pathogenesis of rheumatoid arthritis, and Yiqi Bushen Huoxue decoction can significantly reduce this level, which could be one of the mechanism of Yiqi Bushen Huoxue decoction in the treatment of rheumatoid arthritis.
