摘 要:目的:探讨中脑腹侧被盖区在慢性内脏痛调节中的作用。方法:新生期8、10和12天的大鼠进行结直肠扩张刺激来制备慢性内脏痛模型;采用免疫荧光法检测中脑腹侧被盖区内c-fos表达情况和小胶质细胞活化情况;通过利多卡因微量注射中脑腹侧被盖区后观察内脏痛行为学和c-fos的变化情况。结果:与对照组相比:(1)模型大鼠痛阈值降低,腹外斜肌放电和腹壁撤退反射评分显著增加(P<0.05),表明内脏痛模型成功建立;(2)中脑腹侧被盖区c-fos表达增加,神经元兴奋性增高,注射0.5μl 4%利多卡因后,腹壁撤退反射评分降低,痛阈值增高(P<0.05),并且c-fos表达减少;(3)内脏痛模型大鼠小胶质细胞活化,Iba-1表达增多(P<0.05)。结论:中脑腹侧被盖区参与大鼠慢性内脏痛的调节。Objective: To investigate the effect of the ventral tegmental area on the regulation of chronic visceral pain in rats. Methods: The chronic visceral pain model was induced by colorectal distension(CRD) on rats 8, 10 and 12 days after birth. The expression of c-fos and activation of microglia in ventral tegmental area were detected by immunofluorescent staining. The changes of visceral pain behavior and c-fos expression were also observed before and after injection of lidocaine into the ventral tegmental area. Results: Compared with control group:(1) the distention threshold decreased, and the abdominal withdrawal reflex scores and external oblique discharge amplitude increased significantly in CRD group rats(P〈0.05);(2) the expression of c-fos increased in CRD group. After microinjection of lidocaine into the ventral tegmental area, abdominal withdrawal reflex scores significantly decreased and distention threshold increased(P〈0.05). Moreover, the c-fos expression was inhibited by lidocaine(P〈0.05);(3) the microglia were activated and the Iba-1 expression increased in CRD group(P〈0.05). Conclusion: The ventral tegmental area is involved in the formation of chronic visceral pain in rats.
