摘 要:目的:分析红细胞保存液(MAP)悬浮洗涤红细胞在保存期内的质量指标。方法:将悬浮红细胞随机分为3组,A组不洗涤,B组用生理盐水洗涤后用生理盐水混悬,C组用生理盐水洗涤后用MAP混悬。B组在制备当天取样,A组、C组在制备当天、保存7、14、21、28、35d取样,检测血红蛋白含量、上清蛋白质含量、溶血率及无菌试验。结果:与制备当天相比,悬浮红细胞保存21d后上清蛋白质含量显著升高(P<0.05),保存14d后溶血率显著升高(P<0.05);MAP洗涤红细胞保存14d后上清蛋白质含量、溶血率显著升高(P<0.05)。保存35d的MAP悬浮洗涤红细胞与生理盐水悬浮洗涤红细胞各项指标均符合国家标准,2组的血红蛋白含量差异无统计学意义(P>0.05),C组的上清蛋白质含量、溶血率显著高于B组(P<0.05)。结论:保存期末(35d)的MAP悬浮洗涤红细胞符合质量控制要求,但溶血率和上清蛋白质含量随时间延长而逐渐升高,建议在保存期14d内使用为佳。Objective:To analyze the quality of washed red blood cells(RBCs)suspended in erythrocyte preserve fluid(MAP)during storage period.Method:The suspended RBCs were randomly divided into 3groups,group A was not washed,group B was washed and suspended with physiological saline,and group C was washed with physiological saline and suspended with MAP.Group B was sampled in the preparation day,while group A and group C were sampled in 0d,7d,14 d,21d,28 dand 35dof storage.The content of hemoglobin and the supernatant protein,hemolysis rate were detected and sterility test was done.Result:Compared with the preparation day,in group of suspended RBCs,supernatant protein content increased significantly after 21d(P〈0.05),and the rate of hemolysis increased significantly after 14d(P〈0.05);in group of washed RBCs suspended in MAP,supernatant protein content and hemolysis rate increased significantly after 14d(P〈0.05).Four indicators of washed RBCs suspended in MAP at 35 dand washed RBCs suspended in physiological saline were both in line with national standards,and there was no significant difference of hemoglobin content between the two groups(P〉0.05),however supernatant protein content and hemolysis rate were significant higher in MAP group than those in the saline group.Conclusion:Washed RBCs suspended in MAP at 35 dcould meet quality control requirements,but the rate of hemolysis and supernatant protein content increased with time,and it would be better to be used within the storage of 14 d.
